HPLC法测定舒肝益脾合剂中咖啡酸的含量

Determination of Caffeic Acid in Shugan Yipi Mixture by HPLC

摘要: 为建立舒肝益脾合剂中咖啡酸的HPLC分析,采用Dianonsil C18(5μm×250mm×4.6mm)色谱柱,以甲醇—磷酸盐缓冲液(pH值为4.0,甲醇∶磷酸为13∶87)为流动相洗脱,流速为1.0mL/min,检测波长为323nm,柱温为35℃。结果表明,咖啡酸在0.0396~0.7920μg范围内线性关系良好,r=1,平均加样回收率为99.0%(RSD=2.30%)。该方法操作简单,重现性好,结果稳定,可有效地控制舒肝益脾合剂的质量。

Abstract: To develop a quantitative method for determination of the content of caffeic acid in Shugan Yipi mixture,HPLC was made in the study.The separation was performed by using Diamonsil C18 colume (5 μm × 250 mm × 4.6 mm)with a mobile phase consisting of a mixture of methanolphosphoric acid (13∶87,pH:4.0) and column temperature of 35 ℃ and flow rate of 1 mL/min and detective wavelength of 323 nm. The results indicated that the calibration curve is linear within the range from 0.039 6 to 0.792 0 μg (r=1),and the average recovery was 99.0 % with RSD of 2.30 %.The method can be used for the quality control of Shugan Yipi mixture with the advantages of simplicity,reproducibility and stability.
