HPLC-ELSD法测定黄芪精质量标准的研究

Study on Quality Standard of Astragalus Essence Determined by HPLC-ELSD

摘要: 采用HPLC-ELSD法对黄芪精中黄芪甲苷的含量进行分析。色谱柱为AgilentC18(5μm,4.6mm×250mm),柱温为35℃;流动相:乙腈—水(34:66);流速:1.0mL/min;ELSD检测器参数:漂移管温度为65℃;空气压力为30psi。黄芪甲苷在32.12~321.20μg范围内线性关系良好(r=0.9998),平均回收率为99.26%,RSD=2.75%(n=6)。该法简便、准确、重现性好,可作为黄芪精质量控制方法。

Abstract: The content of astragaloside Ⅳ in Astragalus essence was determined by HPLC-ELSD.The chromatography column was Agilent C18(5 μm,4.6 mm×250 mm);the column temperature was 35 ℃;the mobile phase was acetonitrile and water(34:66);the flow rate was 1.0 mL/min;the temperature of drift tube of ELSD detector was 65 ℃,and the air pressure was 30 psi.Results showed that the astragaloside Ⅳ within 32.12-321.20 μg had good linear relationship(r=0.999 8);average recovery was 99.26%,and the RSD was 2.75%(n=6).It is suggested that the method is simple and accurate and has good reproducibility,so it could be used in quality control method of Astragalus essence.
