青春双歧杆菌培养基的优化及冻干保护剂的筛选

Optimization of Bifidobacterium adolescentis Basic Medium and Screening of Freeze-drying Protective Agents

摘要: 针对青春双歧杆菌的营养需要,采用单因素试验对双歧杆菌培养基进行优化,得出最佳的培养基配方为:蔗糖2%、酪蛋白胨1%、大豆蛋白胨1%、酵母浸膏0.7%、低聚木糖0.3%、磷酸二氢钾0.05%、甘蓝汁8%。试验测定了青春双歧杆菌在最优培养基中的生长曲线,并同时测定pH值和吸光值的变化,确定发酵培养的终止时间为14~16h,活菌数可达2.0×109cfu/mL;同时通过单因素正交试验筛选出其最佳冻干保护剂为:海藻糖20%、脱脂奶粉20%、甘油1.0%、蔗糖8%、明胶5%。

Abstract: According to the nutritional needs of Bifidobacterium adolescentis,the medium for Bifidobacterium adolescentis were optimized by single factor experiment. The optimum medium prescription was confirmed as follows:2% sucrose,1% casein peptone, 1% soya peptone, 0.7% yeast extract, 0.3% xylooligosaccharides, 0.05% KH 2 PO 4 , 8% cabbage juice. The growth curve of B. adolescentis in the optimum medium was determined. The changes of pH and absorption value were determined. It was confirmed that the terminal time for the fermentation culture was 14-16 h hours and the number of active bacteria could reach 2.0 ×10 9 cfu/mL. The optimum freeze -drying protective agents were confirmed by single factor test and orthogonal test : 20% trehalose,20% skimmed milk powder,1.0% glycerin,8% sucrose,5% gelatin.
