雏鸡透明骨骼标本的制作及染色对比试验

Preparation of Chicken Transparent Specimen and Comparison of Staining Methods

摘要: 为了进一步改进动物透明骨骼标本的制作技术,制作完整清晰的骨骼标本,采用3种方法制作雏鸡透明骨骼标本。3种试验方法对动物的处死、固定、脱水、透明方法一致,分别用茜素红和阿利新蓝染液双重染色方法、茜素红和甲苯胺蓝染液双重染色方法、茜素红和阿利新蓝染液混合复染法进行染色。结果发现,用茜素红和阿利新蓝染液双重染色方法制作的透明骨骼标本效果最好,阿利新蓝比甲苯胺蓝染色效果好。

Abstract: In order to improve the preparation technology of animal skeleton transparent specimens, 3 methods for preparation of chicken transparent specimen were compared. The 3 methods shared the same process of sacrifice, fix, dehydration and hyalinization; the staining methods were alizarin red and alcian blue double staining, alizarin red and toluidine blue double staining, and alizarin red and alcian blue counterstaining. The best staining method for skeleton transparent specimens preparation was alizarin red and alcian blue double staining method, and alcian blue performed better than toluidine blue.
