Effect of Different Cells Density on the Cryopreservation of Bovine Mammary EpithelialCells

Abstract: In this study,the breast tissue of a healthy Holstein dairy cow was used.Through five-generation purification,the bovine mammary epithelial cells were divided into four groups,according to the cell density(1×104 cells/mL,group 1;1×105 cells/mL,group 2;1×106 cells/mL,group 3;1×107 cells/mL,group 4, respectively)and cryopreservated. One month later the cryopreservated cells were recovered and cell viability was measured.The results showed that the death rate of bovine mammary epithelial cell was significantly lower for group 4 than that of other treatments(P0.01), whereas the reverse result was found for cell adhesion rate among four treatments (P0.05).As compared with other groups,group 3 had the lowest apoptosis rate(P0.05).The cells of group 3 and group 4 basically covered with the bottom of the culture flask and the cells were in good condition at three days after adherence.
