摘要: 研究以蒙古冰草为材料,通过单因子Mg2+、dNTP、Taq酶、引物、DNA试验;2×Taq PCR Master Mix、引物、DNA正交试验建立了蒙古冰草SRAP反应的优化体系。结果表明:最佳反应体系为10uL:2uL 2×Taq PCR Master Mix、0.4umol/L引物、60ng模板DNA。该体系对蒙古冰草扩增效果好,电泳结果显示扩增条带清晰,多态性高,而且操作简单,为SRAP标记技术在蒙古冰草相关研究中的应用提供条件。
Abstract: Taking the seedling of Agropyron mongolicum Keng as tested material,and establish the optimal SRAP system of Agropyron mongolicum Keng by single factor tests of Mg2+、dNTP、Taq、primer、DNA and orthogonal design approaches of 2×Taq PCR Master Mix、primer、DNA.The results indicated that:10uL reaction system is the most suitable system which containing 2uL 2×Taq PCR Master Mix,0.4umol/Lprimer,60ng template DNA.Using Agropyron mongolicum Keng to test this optimized SRAP system,and the electrophoresis results showed that the amplification bands were clear,highly polymorphic and Simple.This work is a foundation for applying SRAP marker to Agropyron mongolicum Keng molecular biology studies.
