高原低氧环境下大鼠AVP水平与高原习服试验研究

Effects of High Altitude Hypoxia on AVP Level and High Altitude Acclimatization in Rats

摘要: [目的]研究高原低氧环境下大鼠血浆精氨酸加压素(AVP)水平变化,探讨其与高原低氧习服的关系。[方法]将50只雄性SD大鼠随机分为5组,分别为24,48,72h和1周、1个月组,即A、B、C、D、E组,同时设立对照组(西安地区,海拔5m)。试验组分别由西安用汽车运至试验地区,A组为耗时24h带到青海格尔木(海拔2700m),B组为经过48h带到西藏唐古拉(海拔5000m),C、D、E组分别经72h、1周和1个月均带到西藏那曲(海拔4500m)地区,并在不同时间点处死取材,应用放射免疫法检测血浆AVP的浓度,并与对照组比较。[结果]血浆AVP浓度在初入高原后呈上升趋势,随后下降,在48h达到最低,后逐渐回升,至1周时已经接近对照组水平,1个月时达到最高水平。[结论]高原慢性低氧环境下,血浆AVP的浓度随着缺氧时间的延长而变化,这可能与机体缺氧程度及高原低氧习服过程有关。

Abstract: [Objective] To observe changes of plasma arginine vasopressin(AVP) levels and its relationship with altitude hypoxic acclimatization in rats under high altitude hypoxia.[Method] Fifty male SD rats were randomly divided into five groups(A,B,C,D and E),and control group(Xi'an region,altitude of 5 m) was also set up.The experimental groups were transported to test area by car.The rats in the group A were taken to Golmud region of Qinghai Province(altitude of 2 700 m) within 24 h.The rats in the group B were taken to Tanggula region of Tibetan area(altitude of 5 000 m) within 48 h.The rats in groups C,D and E were taken to Naqu region of Tibetan area(altitude of 4 500 m) within 72 h,1 week and 1 month,respectively.All the rats were sacrificed at different time points,and plasma AVP levels were determined by radioimmunoassay.The results were compared with those of the control group.[Result] The plasma AVP level increased after they entered the plateau.It then decreased and reached a valley after 48 h.Subsequently,it gradually picked up after 1 week and reached a peak after 1 month.[Conclusion] Under high altitude hypoxia,plasma AVP level changes with prolongation of hypoxia,maybe due to degree of hypoxia and altitude hypoxic acclimatization.
