家蝇胚胎细胞系染色体制作方法

Preparation Methods of the Chromosome of Cell Lines in Musca domestica Embryo

摘要: [目的]探索家蝇胚胎细胞系(MDEC-07114)染色体制作的方法。[方法]以MDEC-07114细胞为材料制备染色体。实验分组:秋水仙素浓度为5、10mg/L组;秋水仙素处理时间为5、6、10h组;预固定组和直接固定组。[结果]秋水仙素浓度为10mg/L、作用时间为5h组,细胞分裂相多,染色体形态好;预固定处理后细胞分散好,染色体形态清晰。[结论]10mg/L秋水仙素作用5h,经预固定处理后制作MDEC-07114细胞染色体效果最佳。

Abstract: [Objective] The research aimed to explore the preparation method of chromosome of cell lines(MDEC-07114) in Musca domestica embryo. [Method] The chromosome was prepared with MDEC-07114 cells as materials. The experimental groups were set up as follows:groups of 5 and 10 mg/L colchicine;groups of treating with colchicine for 5,6 and 10 h,pre-fixative group and the fixed group. [Result] More cell division and good chromosome morphology can be observed in the group of treating with 10 mg/L colchicine for 5 h. And clear and separated chromosome were observed in pre-fixative group. [Conclusion] The prepared chromosome of MDEC-07114 cells after treating with 10 mg/L colchicine for 5 h and pre-fixative treatment had the best effects.
