Isolation, Culture and Biological Characteristics of Min Pig Ear Skin Fibroblasts

摘要: 为了研究民猪耳皮成纤维细胞分离培养方法及生物学特性,以1日龄民猪耳皮组织为试验材料,细胞原代培养采用组织块贴壁法,成纤维细胞纯化采用胰酶消化法和差速贴壁法,建立民猪耳皮组织成纤维细胞系,同时将细胞液氮冷冻长期保存。结果表明,成纤维细胞生长曲线呈“S”形,冻存前后活率均在90%以上。该试验成功建立了民猪成纤维细胞体外分离和培养方法。

Abstract: This study aimed to establish the isolation and culture method of Min Pig ear skin fibroblasts and to under their biological characteristics. To establish a Min Pig ear skin fibroblasts line, the ear skin tissues of 1-day-old Min Pig were cultivated by explant culture technique and the obtained fibroblasts were purified by enzyme digestion and differential adhesion method. The harvested cells were cryopreserved in liquid nitrogen. The fibroblasts showed a typical "S" shape growth curve and their survival rate before and after freezing were above 90%. The in vitro isolation and culture method of Min Pig ear skin fibroblasts was successfully established in this study.
