不同脱除卵丘细胞方式对牛体外胚胎生产效果的影响

Effect of Different Cumulus Cell Removal Methods on Bovine in Vitro Embryo Production

摘要: 为了提高牛体外受精性控胚胎发育率,试验从体外受精(IVF)培养8 h后的胚胎,采用不同的脱除卵丘细胞方式(机械脱除法、酶脱除法、振荡法)脱除卵丘细胞,研究影响牛性控精液体外受精效率的关键技术。结果表明,在体外受精8 h后,采用不同的脱除卵丘细胞方式(机械脱除法、酶脱除法、振荡法)脱除卵丘细胞,受精卵经体外培养48 h后统计胚胎卵裂率分别为(58.8±2.8)%、(58.5±5.7)%、(69.9±3.4)%,体外培养192 h统计囊胚率分别为(26.7±2.7)%、(26.9±3.9)%、(35.8±4.1)%,由此可见,振荡法显著优于机械脱除法和酶脱除法(P〈0.05)。性控精液体外受精时,体外受精8 h后采用振荡法脱除卵丘细胞对于提高性控精液体外受精胚胎发育率具有重要意义。

Abstract: In an attempt to improve the developmental rate of bovine in vitro fertilized sexed embryo, the embryos cultured in vitro for 8 h were used as material to assess the effect of different cumulus cell removal methods (mechanical removal method, enzyme removal method, oscillation removal method) on efficiency of in vitro fertilization (IVF) of sex-sorted semen. The results revealed that the cleavage rates of the fertilized eggs treated with the three cumulus cell removal modes after 48 h in vitro culture were (58.8±2.8)% ,(58.5±5.7)% and (69.9±3.4)%, respectively, and the blastocyst rates after 192 h in vitro culture were (26.7±2.7)%,(26.9±3.9)% and (35.8±4.1)%, respectively. It was demonstrated that the oscillation removal method had significantly better performance compared to the other two cumulus cell removal methods (P〈0.05). In conclusion, when the sex-sorted semen is used to produce bovine in vitro fertilized embryo, the removal of cumulus cell by oscillation method from the embryos after 8 h in vitro culture can significantly improve the developmental rate of bovine in vitro fertilized sexed embryo.
