紫花苜蓿无菌苗培养前的种子消毒技术研究

Study on The Disinfection Techniques of Seeds for Preparation of Free-germ Seedling of Alfalfa

摘要: 经过不同消毒处理的苜蓿种子的消毒效果表明:种子在消毒前,经无菌水浸泡24h(预处理)比不浸泡的消毒效果好,但是浸泡后,用酒精消毒时间不宜过长;2%次氯酸钠、10%次氯酸钙、0.1%升汞3种消毒剂中.0.1升汞消毒效果最好,种子带菌率为0,用75%的乙醇预处理可大幅度降低种子的带菌率:3种消毒剂中0.1%升汞处理的苜蓿种子发芽率较高。

Abstract: The result of disinfection indicated that the disinfection effect with pre-treatment of soaking in sterile water was better than that of non-treatment, but the disinfection shouldn't be long after treated with soaking, the germ-carrying rate of seed was decreased greatly with pre-treatment of 75 % ethyl alcohol. Among 2 % Sodium Hypochlorite,10 % Calcium Hypochlorite and 0.1% Mercuric Chloride, 0.1% Mercuric Chloride was the best one, the germ-can'ying rate of seed was 0 % after disinfected with 0.1% Mercuric Chloride, and the germination rate of seed also was the highest one.
