骆驼瘤胃内优势纤维素降解菌酶活种类及活力测定方法比较研究

Enzyme Kinds of Dominant Cellulose-degrading Bacteria in Camel Rumen and Detection Methods Comparison of Its Activity

摘要: 利用传统纤维素酶活力测定方法---3,5-二硝基水杨酸法(DNS法)和根据试验具体情况自行构建的纤维素酶活力测定方法,对健康骆驼瘤胃内自行分离的7株优势纤维素降解菌的纤维素酶酶活种类及活力进行了测定。测定结果为:DNS法中不同菌株的CX酶酶活大小为1#>2#>17#>10#>15#>8#>20#;C1酶酶活大小为1#>2#>17#>20#>10#>15#>8#;β-葡萄糖苷酶酶活大小为1#>2#>17#>10#>20#>8#>15#;总酶活大小为1#>2#>17#>8#>10#>15#>20#。自行构建方法中CX酶酶活大小为(1#、2#、15#)>(10#、17#)>(8#、20#);C1酶酶活大小为(1#、2#、15#、17#)>(8#、10#)>20#;β-葡萄糖苷酶酶活大小为(1#、2#、8#、10#、15#、17#)>20#;总酶活大小为2#>1#>15#>17#>10#>8#>20#。根据测定结果可以看出,2种方法所测定的酶活大小趋势较为接近,但在试验过程中DNS法出现诸多问题,弊端较多,而自行构建的方法能够克服DNS法的缺点,使其结果更为简便直观,如果结合DNS法试验数据,则可以使试验结果更加精确。

Abstract: The kinds and activity of cellulase of 7 strains of dominant cellulose-degrading bacteria separated from healthy camel rumen were determined by using traditional determination method of cellulase activity 3, 5-dinitrosalicylic acid method (DNS) and self-constructed determination method of cellulase activity. The determination results showed that the activity of CX in different strains determined by DNS was 1#〉2#〉17#〉10#〉15#〉8#〉20#. And the activity of C1 was 1#〉2#〉17#〉20#〉10#〉15#〉8#. The activity ofβ-glucosidase was 1#〉2#〉17#〉10#〉20#〉8#〉15#. Total enzyme activity was 1#〉2#〉17#〉8#〉10#〉15#〉20#. It was seen from the determination results by the self-constructed method that the activity of CX in different strains was (1#、2#、15#)〉(10#、17#)〉(8#、20#). And the activity of C1 was (1#、2#、15#、17#)〉(8#、10#)〉20#. The activity of β-glucosidase was (1#、2#、8#、10#、15#、17#)〉20#. Total enzyme activity was 2#〉1#〉15#〉17#〉10#〉8#〉20#. In conclusion, the enzyme activity determined by two methods was adjacent. But there were a great number of problems in the determination process by DNS method. And the self-constructed method could conquer the disadvantages of DNS method. The determination results of self-constructed method were simple and intuitive. And the determination results would be more accurate when the self-constructed method was combined with DNS method.
