Maturation in vitro and Parthenogenetic Activation of Mice Oocytes

Abstract: [Objective]The research aimed to used the maturation in vitro of mice oocytes and three activation methods (ethanol activation, strontium chloride activation, ethanol-strontium chloride activation) and make the parthenogenetic activation study of mice oocytes. [ Method ] After mice were injected with pregnant mare serum gonadotropin (PMSG) 48 h, ovaries were picked to obtain three classes of immature oocytes. The immature oocytes of mice were in vitro matured and cultured in maturation culture solution ( 100 mL, M199 +2.2 mg sodium pyruvate +100 IU/mL double antibody + 2 IU/mL LH + 10%FCS). The obtained mature oocytes were activated hy ethanol, strontium chloride, ethanol-strontium chloride. [Result]The maturation rate of A-class oocytes, B-class oocytes and C-class oocytes were 79%, 70% and 55% respectively. The activation rate of A-class oocytes by strontium ch!oride reached 80.0% and this activation method was the best. [Conclusion]The maturation and cuhure effects of Aclass oocytes were the best. The parthenogenetic activation effect of strontium chloride was higher than that of ethanol.
