Abstract: [Objective]To investigate the prevalence of Cryptosporidium infection in primates in Hefei Wildlife Park.[Method]The feces samples were collected from Lemur catta,Erythrocebus patas,Papio hamadryas and the other three kinds of primates reared in Hefei Wildlife Park,in which the presence of oocysts were screened by saturated sucrose solution floating method.The oocysts were stained by acid-fast staining method and were morphologically observed.Furthermore,the genomic DNA of oocysts recovered from the feces samples were extracted and the Cryptosporidium oocyst wall protein(COWP)gene were amplified using PCR assay.The PCR product were subsequently identified by 1%agarose gel electrophoresis.The incidence of Cryptosporidium infection in primates in this wildlife park was calculated.[Results]The morphological identification demonstrated that the size of the obtained oocysts was 4.66μm×5.18μm which was similar to the morphological characteristics of the Cryptosporidium oocysts reported previously;the size of the PCR product was 377 bp which was consistent with the expected size of the COWP fragment;the incidence of Cryptosporidium infection in primates in this wildlife park was 16.67%.[Conclusion]The prevalence of Cryptosporidium infection in primates in Hefei Wildlife Park was observed,indicating a potential zoonotic risk.
