牡丹组织培养繁殖技术初探

Study on the Propagation Technique of Peony in Vitro Culture

摘要: 以“洛阳红”牡丹组培苗为试材,探讨了几种基本培养基、激素配比、培养条件对牡丹继代苗增殖生长的影响。结果表明:DKW基本培养基适宜“洛阳红”牡丹继代增殖,分化数为3.14个,株;WPN培养基次之为3.02个,株,培养基中附加2.0—3.0mg/L 6-BM/与0.5mg/L IAA或0.05mg/LNAA配比有利于继代苗分化、生长;24℃和30℃对“洛阳红”继代苗增殖影响不大,20℃稍次,但30℃下试管苗长势慢、老化很快不宜采用。

Abstract: Tree peony cultured in vitro was studied for elect of basic media,combination of plant growth regulators,incubation condition on subculture status.The results showed that DKW medium and WPM medium were suitable basic medium for Tree Peony' s subculture. Suppling of 2.0-3.0mg/L BA and 0.5mg/L IAA or 0.05mg/L NAA could benefit the subculture.The plandets incubated at 20-25℃ had no significant difference in terms of the number of propagating buds,but the plantlets were aging and dead faster at 30℃.
