老芒麦×紫芒披碱草杂种F1幼穗再生体系的建立

Establishment of Regeneration System of Hybrid F1 of Elymus sibiricus×E.purpuraristatus

摘要: 以老芒麦×紫芒披碱草种间杂种F1代的幼穗(2~4cm)为外植体材料,以MS为基本培养基,研究了4种不同外源激素组合对幼穗愈伤组织诱导和植株再生能力的影响,筛选出最适杂种F1愈伤组织诱导的培养基为:MS+2,4-D2.0mg/L,诱导率达63.3%;适宜幼苗分化的最佳培养基为:MS+6-BA2.0mg/L+NAA0.4mg/L,分化率为75%;适宜生根的最佳培养基为MS+IBA0.5mg/L,生根率达90%;组培苗经过炼苗移栽得到了完整植株.该再生体系的建立为下一步进行秋水仙碱诱导染色体加倍提供基础,最终克服其不育性,创造饲草新种质.

Abstract: In this study, with the young spikes (2-3cm) of hybrid F1 of Elymus scibiricus × E.purpuraristatus as explants and MS as basic medium, the effects of 4 kinds of hormones (2,4-D,6- BA,NAA and IBA) on callus induction, plantlet differentiation and rooting were studied. The optimal medium for callus induction of hybrid F1 was MS+2,4-D 2.0mg/L, with the induction rate of 63.3%;The optimal medium for plantlets differentiation was MS+6-BA 2.0mg/L+NAA 0.4mg/L,with the induction rate of 75%. The optimal medium for rooting was MS+IBA 0.5 mg/L, with the induction rate of 90%;and the regenerated plant was obtained after acclimatization and transplanting. This regeneration system provides conditions for the following colchicin-induced chromosome doubhng, and ultimately overcoming its sterility to create new grass germplasm resource.
