RP-HPLC法检测安替比林血药浓度

Determination of Plasma Antipyrine by Reversed Phase High Performance Liquid Chromatography

摘要: 建立了安替比林血药浓度检测的反相高效液相色谱法(RP-HPLC)。血浆经乙腈提取后进样测定,色谱柱为Kromasil C18(250mm×4.6mm,5μm),柱温为30℃,流动相为0.025mol/L醋酸钠溶液-乙腈(74∶26,V/V),流速为0.8mL/min,检测波长为254nm,进样量20μL。安替比林血药浓度在0.1~30μg/mL范围内线性关系良好(r=0.9995),平均回收率为93.44%~97.21%,日内和日间变异系数小于4.26%。

Abstract: A reversed phase high performance liquid chromatography method(RP-HPLC) was developed for determination of plasma antipyrine.Plasma samples were prepared with acetonitrile.The chromatographic separation was performed on a Kromasil C18 column(250.0 mm × 4.6 mm,5 μm) at column temperature of 30 °C.The mobile phase,a mixture of 0.025 mol/L aqueous solution of sodium acetate and acetonitrile(74:26;V/V),was delivered at a flow rate of 0.8 mL/min.The detection wavelength was 254 nm.The injection volume was 20 μL.Excellent line relationship was obtained in the range of 0.1 to 30 μg/mL(r = 0.999 5).The average recovery rate was 93.44%-97.21%.The intra-day and inter-day RSD were less than 4.26%.
