Research on Tissue Culture and Rapid Propagation of Spiranthes sinensis

Abstract: Using fleshy root, young leaf and rachis of wild flower Spiranthes sinensis as explant, the rapid propagation was constructed with tissue culture in the study. The results showed the induction rate with using rachis as callus reached 83 %, the medium compositing of 3.0 %MS sucrose, 0.75 % agar, 1.0 mg/L 6-BA, 0.2 mg/L NAA and 100 g/L coconut juice was the optimization to inducing, but the optimization medium on proliferation was cumposited of 3.0 % MS sucrose, 0.75 % agar, 2.0 mg/L 6-BA and 0.l mg/L NAA. The optimization of rooting medium was composited of 2.0 % MS sucrose, 0.50 % agar, 0.5 mg/L 6-BA, 0.1 mg/L NAA and 0.5 g/L activated carbon.
