Screening of Teicoplanin-producing Strains Mutation with the Resistance to Self-secondary Metabolites

Abstract: [Objective] Taking teicoplanin product itself as the selective agent,rational screening was made on the producing strain of teicoplanin Actinoplanes teichomyceticus. [Method] The spore suspension of parent strains were irradiated with ultraviolet radiation for certain time and smeared on the isolation medium containing certain teicoplanin product. After culturing for certain time,the grown bacterial colony was selected and the fermentation level was determined. [Result] The fermentation level of the obtained strains was 20% higher than that of parent strains. The subculture test indicated that the obtained strains had higher hereditary stability. The application of the mutant strain in a 50 L fermentor showed a 30% higher fermentation level than parent strain. [Conclusion] It is feasible to make rational screening by using teicoplanin itself as selective agent.
