  • Preparation Methods of the Chromosome of Cell Lines in Musca domestica Embryo

    [Objective] The research aimed to explore the preparation method of chromosome of cell lines(MDEC-07114) in Musca domestica embryo. [Method] The chromosome was prepared with MDEC-07114 cells as materials. The experimental groups were set up as follows:groups of 5 and 10 mg/L colchicine;groups of treating with colchicine for 5,6 and 10 h,pre-fixative group and the fixed group. [Result] More cell division and good chromosome morphology can be observed in the group of treating with 10 mg/L colchicine for 5 h. And clear and separated chromosome were observed in pre-fixative group. [Conclusion] The prepared chromosome of MDEC-07114 cells after treating with 10 mg/L colchicine for 5 h and pre-fixative treatment had the best effects.
    Effects of Different Feeding Methods on the Production Performance and Fat Deposition of Tibet Chicken

    The effects of different feeding methods on the production performance and fat deposition of Tibet chicken were discussed. 120 healthy commercial chicken were selected and fed according to the conventional feeding and management regulations. And the chicken were divided into two groups,one of which was raised in cage and another in freedom,with five repetitions in each group and with 12 chicken in each repetition. The results showed that the feeding methods had significant effects on the body weight and daily weight gain of Tibet chicken. The body weight and daily weight gain of both male and female Tibet chicken in cage were significantly higher than randomly-rearing chicken(P0.05) . The intermuscular fat,subcutaneous fat,the percentage of abdominal fat,the content of intramuscular fat in cage-rearing chicken were significantly higher than that of randomly-raised chicken(P0.05).
    Study on the Physiological Indices in the Serum of Male Sika Deer on Riyue Mountain

    Seven serum physiological indices of ten male sika deer by fence grazing in deer farm on Riyue Mountain were determined and they were compared with some physiological indices of Qilian white-lipped deer,Qilian red deer and Qilian sika deer. The results showed that total albumen in the serum of sika deer on Riyue Mountain was significantly lower than that of Qilian white-lipped deer,Qilian red deer and Qilian sika deer(P0.01) .The inorganic phosphorus in the serum of sika deer on Riyue Mountain was significantly lower than that of Qilian white-lipped deer(P0.01) . And other serum physiological indices had no significant difference(P 0.05).

    The pregnancy rate of frozen semen is easily affected by various factors,such as higher acrosome abnormity rate. The phospholipids and cholesterol in semen might exchange with phospholipids and cholesterol in the diluent,which would decrease the content of cholesterol in the semen and advance the semen capacitation and lead to acrosome reaction. The changes of phospholipid and cholesterol contents during the semen freezing process of goat were systemically analyzed.

    In order to investigate the safety of Bdellovibrio bacteriovorus BD57 to mice,the acute toxicity test was made on mice according to the demands of Veterinary Drug Test Regulation Assemble. The intragastric infusion method and intraperitoneal injection method used in the acute toxicity test. The test results showed that Bdellovibrio bacteriovorus BD57 had no toxicity to mice.

    The minimal inhibitory concentration of eight kinds of botanical natural bacteriostats(such as Syringa vulgaris) to Escherichia coli,Staphylococcus aureus,Candida albicans,Mucor racemose was determined by using tube double dilution method and bacteriostatic test method. The results showed that the inhibitory concentration of Folium Isatidis was the lowest among eight kinds of botanical natural bacteriostats(such as S. vulgaris) ,so Folium Isatidis was the preservative with the best bacteriostatic effect.
    Effects of the First-mating Day on the Farrowing Performance of Sows

    According to the production data of 203 eliminated sows in a pig farm,the litter size at the first parity,parity number,total litter size of sows with different first-mating day were analyzed. The results showed that the alive litter size at the first parity of sows with the first-mating day of 240-269 day-old was the least,the average parity number of sows with the first-mating day of 270-299 day-old was the least,the average parity number and the total alive litter size of sows with the first-mating day of 300-329 day-old were the highest. The average parity number of sows with the first-mating day of 210-239 day-old was the least. To the best economic benefits,the best first-mating day of sows was 210-239 day-old.

    To exhibit the progress of preparing microbial testing slip,several materials such as filter paper,micro-filtration membrane,unwoven fabric or several cold water gelling agents and their combinability were used as the carrier of culture medium to develop microbial testing slips. And their effect was evaluated with Bacillus subtilis and coliforms. It was found that excellent testing slip could be prepared by using micro-filtration membrane with other materials followed with the adjustment of culture concentration and gum dosage,or the combinability of several suitable cold water gelling agents followed with their carrier stacked together face to face. Detectable rate of the ultimate products was above 85%.

    The ecological culture experiment of pond shrimp and crab was made by using micropore pipeline aeration technology. The results showed that 2 800 kg Eriocheir sinensis were harvested in 16 675 m2 pond. And the average unit yield was 112 kg/667 m2 and the average benefits reached 7 500 yuan/667 m2. The micropore pipeline aeration technology had obvious promotion effects on the increase of yield and effeicency of shrimp and crab culture.

The teachers in Jilin University,as a research-university,must focus on the key issue that how to build veterinary medicine specialty well to make it more useful in clinical application and more competitive in basic research. In order to meet the need of research-university,the research aspects of basic veterinary medicine specialty were optimized and integrated into four characteristic aspects like stransgenic animal,molecular pathology and animal models for animal serious epidemic disease,regulation of animal growth and development,and molecular pharmacology of antibacterial agents. The position and research contents of basic veterinary medicine specialty were explored,furthermore the reforming experiences of integrated professional teaching and related experiments were discussed.

    Eight beef cattles were divided into two groups,and they were fed with the dietary containing concentrate and corn cobs. The concentrate contained different protein source,among them DDGS was tested group and cottonseed meal was control group. The apparent digestibility of dietary nutrients was determined through digestion test by total feces collection method. Results showed that apparent digestibilities in total tract of organic matter,crude protein,neutral and acid detergent fiber were unaffected by protein sources significantly. The variation ranges of apparent digestibilities of organic matter,crude protein,neutral and acid detergent fiber in beef cattle ration were 65%~71%,60%~70%,68%~70% and 63%~65%,respectively.

    Effects of Micro-ecological Agent Feed on the Reproductive Performance of Northern Shaanxi White Cashmere Goat

    In order to study the effects of micro-ecological agent EM on the reproductive performance of Northern Shaanxi white cashmere goat,160 healthy 3.5-year-old ewes were divided into two groups,furthermore there was no significant difference in body weight of the two groups(P0.05) ,among them the tested group was fed with crushed dry corn straws which had been fermented by EM fluid,and the control was just fed with crushed dry corn straws without any treatment. Otherwise,all the basal diet was provided by Dingbian Sheep Farm of Shaanxi Province. After six months,oestrus rates of tested group and control group were 93.75% and 87.50%,respectively. Conception rates of tested group and control group were respectively 90.00% and 81.25%,among them the conception rate of tested group was 9.75% more than control. Lambing rates of tested group and control group were respectively 129.16% and 121.54%,while double-lamb rates were 21.51% and 17.72%,and the tested group was 14 lambs more than control. 92 lambs survived in the tested group,while 76 lambs survived in the control group,so the survival rate of tested group was 3.92% more than control,furthermore there was very significant difference in above indexes between tested group and control group(P0.01) . It is concluded that the oestrus rate,conception rate,lambing rate and survival rate of ewes could be improved obviously after they are fed with dry corn straws which had been fermented by EM fluid.

    Cottonseed cake as one kind of high-quality and cheap animal feed protein has been widely applied. But the gossypol in the cottonseed cake limits its dose to some extent. It animals were fed with gossypol-contained feed for a long time,the reproductive toxicity is easily caused. The research situations on the physical and chemical properties,limited amount of gossypol,and its reproductive toxicity to animals were reviewed.

    The effects of various alkali-hydro agents and alkali-hydrolyzable conditions on the determination of tryptophan in the forage feather peptide powder were compared. The results showed that the effect of hydrolyzing with 4 mol/L LiOH at(110±1) ℃ for 20 h was the best,the effect of hydrolyzing with 6 mol/L NaOH and 3% SnCl2 at(110±1) ℃ for 20 h was the next. The effects of the dose of alkali-hydro agent,alkali-hydrolyzable temperature,the concentration of atmosphere and ammonia in the alkali-hydrolyzable tube on the determined value of tryptophan in the determination were further discussed.
    Effect of Botanical Phosphorus Intake in the Diet of Simmental Hybrid Replacement Heifers on the Apparent Digestible Phosphorus

    The relationship between botanical phosphorus intake in the diet of Simmental hybrid replacement heifers and the apparent digestible phosphorus. 24 healthy Simmental hybrid replacement heifers were randomly divided into 6 groups,and there were 4 heifers in each group. The concentrate diet with 6 different levels of phosphorus was fed to each heifer according to 3.50 kg/d,while crude fodder was fed freely. The dietary P content was 0.36%,0.40%,0.42%,0.46%,0.50% and 0.54% on a dry matter(DM) basis,respectively. The total experimental period was 30 d;the normal test comes after 20 d raise period,and fecal samples and urine samples were collected during the last 7 days. The contents of phosphorus and dry matter in diet and fecal samples were determined,and then the regression relationship between phosphorus excretion in fecal samples and botanical phosphorus intake in diet. Results showed that the apparent digestible phosphorus in digestive tract were significantly different(P0.05) from the different treatment of dietary phosphorus intake. To increase dietary phosphorus would be increased fecal phosphorus excretion. However,the apparent digestible phosphorus in digestive tract possessed a parabola function relation with phosphorus intake. The maximum amount of apparent digestion and apparent digestibility in the total tract were 10.5 g/d and 40.8%,respectively.

    The ecosystem service value and functional value of different grassland types in Dulan County of Qinghai Province were evaluated. The results showed that the annual average value of grassland system was 55.40 ×108 RMB yuan. And the total ecosystem service value provided by various kinds of grassland were as follows:temperate desert(16.50×108 RMB yuan) temperate grassland(13.64×108 RMB yuan) alpine meadow(11.60×108 RMB yuan) alpine grassland(7.61×108 RMB yuan) lowland meadow(6.05×108 RMB yuan) . Among various kinds of functional value of grassland,the economic value for providing the food production was only small part of grassland service value. The main service for human provided by grassland ecosystem was its potential indirect ecosystem function and service value.
    Investigation on the Change Patten of Total Volatile Basic Nitrogen in Beef During Storage

    Carcass at 24 h after slaughter were divided into 2 groups and stored at cutting and deboning room and acid discharge room respectively,and then the content of total volatile basic nitrogen(TVB-N) in beef samples was determined every 12 h. Results showed that the content of TVB-N increased with the increase of storage time,and there was obvious difference in TVB-N under different storage conditions,among them the TVB-N of beef stored at cutting and deboning room was obviously higher than that at acid discharge room. The beef stored at cutting and deboning room for 60 h exceeded the standard(15 mg/100 g) ,while that stored at acid discharge room for 108 h exceeded the standard.

    In order to study the effects of vitamin on the reproductive capacity of breeding rabbits,the newly-designed formula feed and original conventional formula feed were fed to rabbits under the same conditions in the test. The effects of vitamin on the oestrus time of breeding rabbits,pregnancy rate,parity number,litter size,survival rate of weaned young rabbits were analyzed. 100 healthy breeding rabbits with the same reproductive age were selected and divided into two groups(group A and B) ,with 50 rabbits in each group(male∶ female=1∶4) . The rabbits in group A were fed with the conventional feed and that in group B were fed with experimental feed. The results showed that vitamin can significantly increase the reproductive capacity of breeding rabbits(P0.01) . During four test months,the estrous female rabbits in group B was 2.46 times as that in group A. The average pregnancy rate of female rabbits in group B was 1.72 times as that in group A. The litter size of female rabbits in group B was 11.58 times as that in group A. The survival rate of young rabbits in group B was twice as that in group A. And the ideal mating and reproduction results were obtained.
    Screening of Teicoplanin-producing Strains Mutation with the Resistance to Self-secondary Metabolites

    [Objective] Taking teicoplanin product itself as the selective agent,rational screening was made on the producing strain of teicoplanin Actinoplanes teichomyceticus. [Method] The spore suspension of parent strains were irradiated with ultraviolet radiation for certain time and smeared on the isolation medium containing certain teicoplanin product. After culturing for certain time,the grown bacterial colony was selected and the fermentation level was determined. [Result] The fermentation level of the obtained strains was 20% higher than that of parent strains. The subculture test indicated that the obtained strains had higher hereditary stability. The application of the mutant strain in a 50 L fermentor showed a 30% higher fermentation level than parent strain. [Conclusion] It is feasible to make rational screening by using teicoplanin itself as selective agent.
