Immunohistochemistry and Image Analysis on Cashmere Goat Mast Cells Tryptase

Abstract: [Objective] Whether there was tryptase in the mast cell of cashmere goat and effects of different stationary liquids on its normal dyeing and immunohistochemistry dyeing result were studied. [Method] Tryptase existing in some mast cells like jejunum, omasum and lung tissue of cashmere goats which were fixed by Carnoy liquid and neutral formalin (NBF) were determined by the rabbits of mast cell tryptase polyclonal antibody indirect immunoperoxidase technique. Experimental results were analyzed by using Image-Pro Plus image analysis software and artificial counting method, and the morphology and amount of mast cells in different fixed tissues after normal dyeing and immunohistochemistry dyeing were compared so as to judge the effects of Carnoy liquid and NBF on the technique. [Result] The polyclonal antibody β2 and cashmere goat mast cell tryptase could obtain good cross reaction, which suggested that there was tryptase in the mast cell of cashmere goat cytoplasmic granules. Compared with Carnoy liquid, NBF-fixed tissue could reflect the morphology and amount change of mast cells in different fixed tissues effectively. [Conclusion] There are tryptase in the mast cell of cashmere goat cytoplasmic granules. Compared with Carnoy liquid, NBF is more suitable for the normal dyeing and immunohistochemistry dyeing of cashmere goat mast cell.
