Comparison of Methods for Detecting Aflatoxins B1 in the Feedstuffs by HPLC and LC-MS/MS

Abstract: In order to provide reference for the selection of appropriate methods for aflatoxin B1(AFB1) detection in laboratory, the detection performance of AFB1 in feedstuffs by HPLC with photochemical derivatization, HPLC with trifluoroacetic acid derivatization and LC-MS/MS were verified and compared in this study. The results showed that HPLC with photochemical derivatization and HPLC with trifluoroacetic acid derivatization had good linear range and correlation coefficient (R≥0.999 97), the recovery rates of AFB1 ranged from 88.4% to 104.6%, and the precisions were below 5%; the correlation coefficient (R) of LC-MS/MS method was ≥0.999 00, the recovery rates of AFB1 ranged from 85.3% to 96.8%, and the precisions were below 3%. The combined data suggest that for the determination of AFB1 in feedstuffs, HPLC with photochemical derivatization has the advantages of simplicity of operation, high sensitivity and good stability, but had higher cost; HPLC with trifluoroacetic acid derivatization has lower cost, but is susceptible to the interferences from matrix in sample, and the operating procedures are complicated; LC-MS/MS has higher qualitative accuracy, but has lower quantitative accuracy and the highest cost in the three methods.
