Establishment of Type 2 Diabetes Mellitus Animal Model of SD Rats and Expression of Pancreatic Tissue SUR1 mRNA

Abstract: [Objective] The research aimed to establish a rat model for type 2 diabetes mellitus(T2DM)induced by streptozotocin(STZ)in combination with high-sugar and high-fat diet and detect the changes of weight,glucose,insulin,insulin sensitivity index, mRNA expression of pancreatic tissue SUR1. [Method] 30 healthy male SD rats were randomly divided into model group(20 rats )and blank group(10 rats). After rats in model group was fed with high-sucrose and high-fat diet for 4 weeks, 30 mg STZ was injected by left- lower abdominal cavity injection per one kilogram body weight. The fasting body weight, blood glucose, insulin, insulin sensitivity index(ISI) were detected on the 1st,4th,8th,12th week after injecting STZ for statistical analysis. After rats were killed, pancreas was picked and the expression of pancreatic tissue SUR1 mRNA by using RT-PCR method. [Result] The success rate of T2DM rat model was 75%. On the 1st, 4th, 8th and 12th week after STZ injection, blood glucose in model group significantly increased than blank group(P0.01). ISI in model group significantly decreased than blank group (P0.01). The expression of SUR1 mRNA in model group was significantly lower than blank group(P0.05). [Conclusion] T2DM animal model could be induced by high-sugar and high-fat diet and STZ left- lower abdominal cavity injection. The decrease of SUR1 mRNA might be one of molecular pathogenesis of diabetes.
