  • Isolation,Identification and Biological Characteristics of Lactic Acid Bacteria

    One strain W7 with high acid production was obtained from the intestinal of chicken and pickle by using the conventional microbiology method. The strain W7 was identified by morphological observation, physiological biochemical test and 16S rRNA gene sequence analysis, and biological characteristics of the strain W7 was studied. Results showed that the strain W7 was identified as Lactobacillus plantarum. The amount of W7 reached the maximum at 37 ℃ for 22 h. The strain W7 had strong resistance to acid, and the survival rate still reached 100% at pH 2.0. The strain W7 could grow when the concentration of bile salt was 0.30%. The strain had good antibacterial effect on some pathogenic bacteria, and the adhesion rate was 79.9%.
    Effects of Stratiomyiid Larva on the Growth Performance and Blood Indices of Broilers

    Four hundred 70-day-old typical Cenxi three-yellow chicken were randomly divided into four groups with 100 chicken in each group. Chicken in control group, group Ⅰ,Ⅱ,Ⅲ were fed with basal diet(2.0% fishmeal ), the diets with 2.0%,4.0%,6.0% Ptecticus aurifer larva powder respectively. The test lasted for 28 days.The effects of replacing fishmeal by P. aurifer larva powder on the growth performance,serum biochemical indices, muscle amino acid content and fatty acid content were investigated. The results showed that 2.0% P. aurifer larva powder could promote the growth, accelerate the growth and development of immune organs and enhance the contents of total amino acids and saturated fatty acids in the muscles. In the group of adding 2.0% P. aurifer larva powder, the serum physiological indices increased except for blood urea nitrogen.
    Study on the Fermentation Conditions of Antibacterial Substance Produced by Bacillus subtilis and the Stability of Its Metabolic Products

    Bacillus subtilis L7 that was isolated from cow dung and the antibacterial substance in its metabolites had strong inhibition on multi-pathogenic bacteria. The studies on the fermentation conditions for producing the antibacterial substance showed that the optimal fermentation conditions were as follows:pH of 7.0, culturing with 4% inoculum amount under constant-temperature oscillation incubator for 60 h at 30 ℃. The antibacterial substance in its metabolites was stable at 80~ 121 ℃ for 30 min under pH of 6.0~10.0. The antibacterial activity decreased under illumination conditions after 2~6 h. In conclusion, the fermented products of strain L7 had stronger stability, so it could be preserved under the environment of lucifuge,neutral pH and room temperature for a short term.And it might be used as a new kind of natural antiseptic in food industry or applied in the production of anti-infection micro -ecological preparation after treatments of increasing the activity and purifying.
    Establishment of Type 2 Diabetes Mellitus Animal Model of SD Rats and Expression of Pancreatic Tissue SUR1 mRNA

    [Objective] The research aimed to establish a rat model for type 2 diabetes mellitus(T2DM)induced by streptozotocin(STZ)in combination with high-sugar and high-fat diet and detect the changes of weight,glucose,insulin,insulin sensitivity index, mRNA expression of pancreatic tissue SUR1. [Method] 30 healthy male SD rats were randomly divided into model group(20 rats )and blank group(10 rats). After rats in model group was fed with high-sucrose and high-fat diet for 4 weeks, 30 mg STZ was injected by left- lower abdominal cavity injection per one kilogram body weight. The fasting body weight, blood glucose, insulin, insulin sensitivity index(ISI) were detected on the 1st,4th,8th,12th week after injecting STZ for statistical analysis. After rats were killed, pancreas was picked and the expression of pancreatic tissue SUR1 mRNA by using RT-PCR method. [Result] The success rate of T2DM rat model was 75%. On the 1st, 4th, 8th and 12th week after STZ injection, blood glucose in model group significantly increased than blank group(P0.01). ISI in model group significantly decreased than blank group (P0.01). The expression of SUR1 mRNA in model group was significantly lower than blank group(P0.05). [Conclusion] T2DM animal model could be induced by high-sugar and high-fat diet and STZ left- lower abdominal cavity injection. The decrease of SUR1 mRNA might be one of molecular pathogenesis of diabetes.
    Optimization of Extraction Process of Baphicanthus Compound Feed Additives by Comprehensive Scoring Method

    [Objective] The aim was to establish an efficient and environmental protection extraction process of the Baphicanthus co-mpound feed additives. [Method] Ultrasonic extraction was used as an assistant method, and then the extraction solvent, extraction time, liquid ratio and extraction times in extraction process of the Baphicanthus compound feed additives were investigated by using L9(34) orthogonal design. The gain ratio of dried extract, total content of adenosine and total content of chlorogenic acid were taken as the quantitative indexes. [Result] The best extraction condition of Baphicanthus compound feed additives was as followed: 16 times amount of 50% ethanol was added and then extracted for two times, with 45 min for each time. [Conclusion] The optimized extraction technology is simple, stable, reproducible and good for large-scale production. The research provides basis for the extraction process of the Baphicanthus compound feed additives.
    Effects of Compound Probiotics and Feed Antibiotics on the Serum Biochemical Indices and Intestinal Enzyme Activity of Broilers

    The effects of adding compound micro-ecological preparation and feed antibiotics on the serum biochemical indices and intestinal enzyme activity of broilers were studied. Five hundred one-day-old female AA broiler chicken were randomly divided into five groups with 4 repetitions in each group and 25 chickens per repetition. The pre-trail period of the experiment was 7 days. The chicken in the control group(groupⅠ)were only fed with the basal diet(no antibiotics),chicken in groupⅤwas fed with antibiotics and chicken in group Ⅱ,Ⅲ and Ⅳ were fed with feeding the diet with adding 1.0‰,2.0‰ and 4.0‰ micro-ecological preparation. The test period lasted for 35 days. The results showed that the content of serum total protein in group Ⅲ in each test stages was significantly higher than that in control group and antibiotics group(P0.05), except 42-day-old. Except 14- day-old, the content of serum urea nitrogen in group Ⅲ of broilers was significantly lower than that in control group and antibiotics group (P0.05). In all test stages, the activity of protease in ileum contents in group Ⅲ was significantly higher than that in control group and antibiotics group (P0.05). And the activity of protease in jejunum contents in group Ⅲ was significantly higher than that in control group and antibiotics group (P0.05), except 14-day-old. Except 14-day-old, the activity of amylase in ileum and jejunum contents in group Ⅲ was significantly higher than that in control group and antibiotics group(P0.05). Therefore, 2.0‰ compound micro-ecological preparation could significantly increase the level of total protein in the serum of broilers, enhance the activities of protease and amylase in ileum and jejunum contents and decrease the content of serum urea nitrogen of broilers to some degree.
    Effect of Cashmere Washing Factors on FMD Virus under Experimental Conditions

    Imitating washing technology of cashmere in Mongolia and China, the inactivating effect of FMD virus in cashmere was detected under different washing temperatures and treating times as well as different drying temperatures and treating times. Results indicated that the FMD virus NM-zg99 strain in the cashmere could not be inactivated by treating in the washing water at below 64 ℃ for 60 min. However, the virus could be inactivated by treating at 65 ℃ for 60 min, 66 ℃ for 45 min, 75 ℃ for 10 min and 80 ℃ for 5 min. There was a close relation between drying temperature and the treating time for the inactivation of the virus in cashmere. The virus could be inactivated by treating at 70 ℃ for 35 min and 75 ℃ for 25 min, 80~90 ℃ for 20 min, 95~105 ℃ for 15 min and 110 ℃ for 10 min. The result provides technical reference for making test standard for importing cashmere washing in China and also has directive significance for the cashmere washing corporations to improve the washing technology of sheep wool and cashmere.

    [Objective] The aim was to compare 4 serology diagnostic tests for brucellosis. [Method] 876 serological samples were detected by rose bengal plane test (RBT), indirect enzyme linked immunosorbent assay (i-ELISA), competitive enzyme linked immunosorbent assay (c-ELISA) and standard agglutination test (SAT). [Result] The number of positive samples detected by RBT, i-ELISA, SAT and c-ELISA were 543, 501, 460 and 500, and the positive rates were 61.99%, 57.19%, 52.51% and 57.08%, respectively.
    Screening of Anti-PRRSV Peptide by Phage Peptide Library and Its Application

    The phage display peptide library is a widely-used epitope identification technology,which is combined with the protein screening. Phage display technique was used to screen out the proteins interacted with ORF1B copy enzyme protein of porcine respiratory and reproductive syndrome virus (PRRSV) . And the antiviral activity of the screening protein was further verified. The purified CTD protein of PRRSV ORF1B that coated high-affinity 96T board was used as the target protein. T7 phage display technology was applied to screen out the cDNA library of random 12-peptide library. And the screened clones were sequenced and analyzed. After the synthesis of the screened clone sequence, the anti-PRRSV effect of the synthetic polypeptide was verified in vitro. The results showed that 87 positive clones were obtained after 4 circles of phage screening. 11 screened 12-peptide were identified after sequencing. Through in vitro antiviral experiment, it was confirmed that P10 was a 12-peptide with high-antiviral activity. These results laid the foundation for the antiviral research of PRRSV.

    Antimicrobial peptides(AMPs),named as peptide antibiotics in another way,are a kind of micromolecular peptides with immune activities that are encoded by special genes,synthesized by ribosome and secreted by some glands. A lot of experiments proved that the antimicrobial peptides not only had broad-spectrum efficient antibacterial activities,but they had stronger killing effect on some fungi,protozoon,virus,tumor cells,etc. And some AMPs even could inhibit the proliferation of HIV-1 virus. The current research situations of antimicrobial peptides were expounded and the application and existing problems were deeply discussed.

    Canine distemper is an acute infectious disease caused by the canine distemper virus in Canidae.     The works of lightning protection and disaster reduction is one of social management functions that should be performed by law for meteorological departments in charge at different levels. As the lightningprooof works gradually perfected, the works of lightning protection and disaster reduction were further strengthened. But the current works of lightning protection and disaster reduction still have such problems as non-observance of related laws and eluding the managements. And the reasons that caused these problems included that incomplete policies and regulations, lack of professionals, less lightningprooof consciousness, lack of government supervision, and so on. Those problems caused that the administrative enforcement of lightningproof works couldn′t went along entirely and effectively. And those problems are the existing hidden troubles in the works of lightning protection and disaster reduction, meanwhile they are urgent difficulties to be solved at present.
