  • Separating and Screening on the Cellulose-decomposing Bacteria and Study on Its Enzyme Activity

    Four strains contained strong degrading ability cellulose-decomposing bacteria were isolated from rotten straw, leaves, soil in corn field, and animal dung from college of animal science and medicine of Inner Mongolia Agricultural University. The diameters of the four strains were up to 3920.8, 808.8, 541.8 and 5110.56 μm respectively after seven days' cultivation. The degradation rate reaches peak value when pH is 6.0 or 6.5, and the highest enzymatic activity can be up to 203, 301,262 and 276 U respectively.
    Determination of Polysaccharides in Tricholoma Mongolicum Mycelium

    The extraction technology of soluble polysaecharides isolated from Tricholoma mongolicum were studied in this paper. Single factor test was applied to analyze the effects of solid-liquid ratio, temperature, time and repeated extract times on the extraction percentage of the soluble polysaccharides. The results indicated that the temperature and solid-liquid ratio were the main factors which 'affect the extraction rate of the soluble polysaccharides.The optimum solid-liquid ratio was1:50, the temperature was 85 , the extraction time was 2 h and was extracted twice.The contents of polysaccharides in the two samples (Tr-Sea and Tr-12) were 6.63 % and 6.40 %, respectively.
    Effects of Urea Concentration on Growth and Fatty Acid Composition of Phaeodactylum tricornutum

    Phaeodactylum tricornutum was cultured in medium with different urea concentration to investigate its growth and fatty aeid composition. The results showed that adding of urea in medium could improve the cell intensity of P. tricornutum markedly. The growth increased with urea concentration rising from 0 to 8.0 mmol/L. The content of eicosapentaenoic acid (EPA) and polyunsaturated fatty acids (PUFAs) increased at first and then dropped with a maximum EPA content (19.1%) at 2.1 mmol/L of urea. The cell intensity was relatively high when 2.1 mmol/L was set as the optimal urea concentration. Considering biomass of algae and production of EPA and PUFAs, this urea concentration is also suitable to get higher production of both.
    The Effect of Gap Junction on Bovine Oocyte Maturation in Vitro

    The effect of gap junction on bovine oocyte maturation and cumulus expansion in vitro was studied by means of treating bovine oocyte with different density of FSH. The results indicated that the optimal density of Follicle-Stimulating Hormone (FSH) was 1.2 IU/mL and the gap junction between oocyte and cumulus cell in vitro had direct influence on bovine oocyte maturation. Furthermore, the gap junction can stimulate bovine oocyte maturation and cumulus expansion in vitro.
    Effects of LDL on Cryopreservation of Sheep Spermatozoa

    The fertility test showed that pregnant rate of AI with frozen semen were significantly lower than that of natural mating. One of the reasons might be the sperm plasma membrane was serious injured during freezing. Therefore, the paper studied on sheep spermatozoa during freezing which was protected with egg yolk (EY) and local fine wool sheep was used as the experimental animals. Then low density lipoprotein (LDL) was used in diluents instead of egg yolk to study on the effect of LDL on sperm motility, acrosome intact rate and plasma membrane integrity. The optimal LDL dilution formula was screened to improve sheep semen quality, reduce the damage sperm, and provide theoretical basis for improving fertility of ram sperm after cryopreservation.
    Analysis on Economic Traits and Candidate Genes of Inner Mongolia White Cashmere Goats

    120 Inner Mongolia cashmere goats were selected in this study. Parts of the sequences of KAP1.3 and KAP6.1 genes from cashmere fiber protein were selected as the candidate genes by PCR-SSCP techniques. The selection of the economic traits of the candidate genes were evaluated through studying on its Polymorphism. The results showed that there was significant correlation between the site S2 and the cashmere yield trait(P〈0.05) in KAP6.1 in the muhi-gene family of the keratin accessory protein by oneway Anova of the analysis software. The AA and BB genotypes were also significant correlated with cashmere yield trait by multiple comparison (P〈0.05). The AB and BB genotypes had signifieant effects on fine wool quality (P〈0.05).
    The Effect of Different Stocking Rates on Grazing Behavior of Sheep in Stipa breviflora Desert Steppe

    The experiment was conducted in stipa breviflora desert steppe in Wang fu number one group, experimental base of Agriculture and Animal Husbandry. Academy, Siziwang banner, Wulanchabu league of Inner Mongolia from 2006 to 2007.Different stocking rates were set up with sheep in order to study grazing behavior of sheep in response to stocking rate. The results showed that the foraging and ruminant behaviors of sheep were significantly affected under different stocking rate. The biting time was obviously reduced while the ruminating time was prolonged when the stocking rate was low. However, the different stocking rates had little impact on other maintenance and excretion behaviors.
    Study on Callus Inducement and Anti-browning of Cyclamen persicum Mill

    The leaves, leafstalk and peduncles of Cyclamen persicum Mill were taken as expand in this experiment, and they were cultured on MS basic medium respectviely with different kind of phytohormones among various concentrations. The influences of phytohormones on different explants and the role of active carbon (AC), vitamin c (VC), polyvinylpyrrolidone (PVP) and darkness culture in anti-browning of explants were studied in this experiment by inducing the callus in different medium. The results showed that the best explants for inducing callus was young leaves, leafstalks and peduncles took the second place. The best medium for callus induction of leaves, leafstalks and peduncles were 1/2MS+6-BA2.0mg/L+2,4-D0.5mg/L+KT0.25mg/L MS+6-BA0.2mg/L+ NAA2.5mg/L, and MS+6-BA0.2mg/L+2,4-D0.5mg/L respectively. Darkness culture was best for anti-browning among the five treatments.
    cDNA of Human Cholesteryl Ester Transfer Protein Cloning and Construction and Identification of Its Eukaryotic Expression Vector

    In order to lay a foundation for further study on the structure and function of CETP, the ORF sequence of human CETP eDNA was cloned and the eukaryotie expression vector was constructed. Total RNA was extracted from human liver tissues and mRNA was purified. Using kit, mRNA was synthesized to cDNA strand through reverse transcription. Primers were designed and the ORF sequence of human CETP cDNA was amplified from synthesized cDNA strand by PCR. And then 1.5 kb eDNA amplified fragment was cloned into pcDNA3.1/myc-His (-)A to form recombinant plasmid vector. Finally, the recombinant plasmid was sequenced after identified. The results showed that the recombinant eukaryotic expression vector contained the complete ORF sequence of CETP eDNA and the sequence was in full accord with the ORF sequence of the reported human CETP eDNA. The eukaryotie expression vector of human cholesteryl ester transfer protein was constructed successfully in this study.
    Effects of Confinedness and Grazing on Immune Function of Cashmere Goats

    By detecting the immune function indices of 30 cashmere goats in the condition of eonfinedness and 30 ones in the condition of grazing,the results showed that comparing with the confinedness group, the positive rate of T-lymphocytes in blood had not significant difference (P〉0.05) in the grazing group. Neutrophilic granulocyte phagocytosis function was significant higher (P〈0.05). The transformation index of lymphocytes was enhanced and there was significant difference (P〈0.05) in the grazing group. In blood, the content of CD4^+ molecule was much higher while the content of CD8^+ had not significant difference. CD4^+/CD8^+ of the grazing group was notably higher (P〈0.01). E-C3 bRR rosette rate and E-ICR rosette rate of grazing group were significant higher (P〈0.05) than the confined group. The leukocyte count, basophile granulocyte numbers and Monocyte numbers of the grazing group were significant different (P〈0.05) compared with the confined group (P〈0.05). The count of segmented and band neutrophilie granulocyte both had notable difference (P〈0.01). Serum total protein and Globulin were significant higher of the grazing group (P〈0.05) except albumin. The content of serum nitric oxide (NO) in grazing group was significant higher than the confined one (P〈O.05). This experiment indicated that the immune function of cashmeres in the condition of grazing were higher than the confined group.
