老芒麦与紫芒披碱草正、反交F1植株同工酶分析

Isozyme Analysis on Reciprocal Hybrids F1 between Elymus sibiricus and Elymus purpuraristatus

摘要: 采用聚丙烯酰胺垂直平板电泳技术对老芒麦与紫芒披碱草及其正、反交F1的过氧化物酶(POD)同工酶、酯酶(EST)同工酶做了比较分析。结果表明,同一生育阶段正、反交F1植株均继承了双亲的基带,表现稳定的遗传特性,同时正、反交F1及亲本在酶带数目、位置方面差异显著,这从酶蛋白分子水平证明正、反交F1的真实性。同工酶酶谱表征可作为正、反交杂种F1与其亲本相互鉴定和识别的依据之一。

Abstract: The proxidase (POD) and esterase (EST) isozyme characteristics of reciprocal hybrids F1 between Elymus sibiricus and E. purpuraristatus were analyzed by vertical slab polyacrylamide gel electrophoresis technology.The results showed that the reciprocal hybrids F1 had all the bands of both parents at the same developmental stage,and there were significant differences in the number, locus and intensity of the POD and EST isozyme bands among the reciprocal hybrids F1 between Elymus sibiricus and E. purpuraristatus.The isozymes provide evidence for the reliability of reciprocal hybrids F1, and the isozymes can be used for identification of reciprocal hybrids F1 and parental lines of Elymus sibiricus and E. purpuraristatus.
