麝香百合组织培养快繁体系建立

Establish of culture rapid propagation system for Lilium longiflorum tissue

摘要: 以麝香百合为研究对象,以MS为基本培养基,优化麝香百合外植体最佳灭菌方式,最终选用5%次氯酸钠浓度,灭菌时间为10min。通过对8种附加不同浓度6-BA,NAA的MS培养基诱导丛生芽及再生植株的研究发现,在MS+1mg/mL6-BA+0.1mg/mLNAA培养基中麝香百合增殖效果良好,增殖系数为6.88。蔗糖浓度为60%时,试管内结鳞茎效果最好,结鳞茎率达到100%,鳞茎直径为1.12cm,质量为2.26g。在1/2MS+0.5mg/mLNAA培养基中进行生根,生根率达到100%,平均根长为2.83cm。最终,对得到的试管苗炼苗移栽,成活率达83%,成功建立麝香百合组织培养技术体系,为赤峰地区的麝香百合组织培养提供一定的技术指导。

Abstract: Lilium longiflorum as the research object, with MS as the basic culture medium,optimize the lilies explant best way to sterilization, eventually choose 5% concentration of sodium hypochlorite, sterilization time for 10 rain. Based on 8 additional different concentrations of 6 - BA, NAA of MS culture medium, according to a study in MS+I mg/mL 6-BA+0.1 mg/mL NAA medium lilies proliferation effect was good, proliferation coefficient of 6.88. Sucrose concentration was 60%, the best bulb effect in vitro, the produce rate of bulbs produce bulbs rate of 100%, bulb was 1.12 cm in diameter, mass was 2.26 g. In 1/2 MS + 0.5 mg/mLNAA medium to take root, rooting rate of 100%, average root length was 2.83 cm. In the end, getting the tube seedling transplanting, the survival rate was 83%.We established Lilium longiflorum system of tissue culture technology and provided certain technical guidance for chifeng area.
