利用流式细胞仪鉴定9个居群蒙古冰草的染色体倍性

Chromosome Ploidy Determination of 9 Populations of Agropyron mongolicum Keng by Flow Cytometry

摘要: 利用3种分离液制备了乌拉尔图小麦细胞核悬浮液,其中Otto缓冲液制备效果最好。选用该分离液制备了9个居群蒙古冰草的单细胞悬浮液,在显微镜下统计细胞数,并利用流式细胞仪检测其染色体倍性,用二倍体乌拉尔图小麦作为对照。结果发现,流式细胞仪检测结果与传统的染色体制片法的结果一致,均为二倍体。

Abstract: Three kinds of separation medium were used to prepare nuclear suspension of Triticum urartu. The preparation effect of Otto buffer was the best. Otto buffer was selected to prepare single cell suspension of 9 populations of Agropyron mongolicum Keng. The number of cells was counted by a microscope and its chromosome ploidy was detected by using flow cytometry (FCM) with T. urartu diploid as control. The results showed that the results detected by flow cytometry were consistent with the results by traditional chromosome preparation method and they were diploid.
