Effects of IBRV Infection on the Nitric Oxide Produced by Bovine Monocyte Macrophage

摘要: 为探讨牛传染性鼻气管炎病毒(IBRV)感染牛单核巨噬细胞后的免疫机理.采用Ficoll—Hypaque密度梯度离心法分离牛外周血单个核细胞,再经贴壁培养获得单核巨噬细胞。应用Griess法检测接种IBRV后6、12、24、48h时间段的牛单核巨噬细胞分泌一氧化氮(NO)的含量,结果显示,单核巨噬细胞感染IBRV后产生NO的量均高于对照组(P〈0.05),攻毒24h时巨噬细胞产生NO的量达到最高水平(607.87μmol/L)。结果表明,IBRV感染可以对单核巨噬细胞分泌NO的量产生影响,这可能和IBRV引起的致病机理有一定关系。

Abstract: To discuss the immune mechanism of infectious bovine rhinotracheitis virus (IBRV) after infecting on bovine monocyte macrophages, bovine peripheral blood mononuclear cells were isolated by Ficoll-Hypaque density gradient centrifugation and monocyte macrophages were obtained by adherent culture. The content of nitric oxide (NO)secreted by bovine monocyte macrophages was detected by Griess method after IBRV was infected 6, 12, 24 and 48 hours. The results showed that the content of nitric oxide (NO) secreted by bovine monocyte macrophages after IBRV was injected was higher than that of control group (P〈 0.05) and the amount of nitric oxide produced by macrophages reached the highest level (607.87μmol/L)after 24 hours. IBRV infection could affect the amount of nitric oxide secreted .by monocyte macrophages,which might be related to pathogenesis caused by IBRV.
