Protective Effect of Echinops latifolius Polysaccharide B against Oxidative Damage in INS-1 Cells

摘要: 旨在探究蓝刺头多糖B(ETPB)对H_2O_2造成的INS-1细胞氧化损伤的保护作用。体外培养INS-1细胞,通过采用MTT法检测细胞存活率,筛选出50μmol/L H_2O_2作用24 h可建立氧化损伤模型,并确定ETPB的最大安全浓度为250μg/mL;ETPB(15.6、31.2、62.5μg/mL)作用48 h后加入H_2O_2作用24 h,可显著提高细胞存活率、SOD活性,降低MDA含量(P〈0.05或P〈0.01)。结果显示,ETPB对由H_2O_2引起的INS-1细胞氧化损伤具有保护作用。

Abstract: This study aimed to assess the protective effect of Echinops latifolius polysaccharide B (ETPB) against oxidative damage in INS-1 cells induced by H2 O2 . The cultured INS-1 cells were used to prepare an in vitro oxidative damage model with treatment of 50 滋 mol/L H2 O2 for 24 hours by determination of cells viability using MTT method; the maximum safety concentration of ETPB for INS-1 cells was determined as 250 滋 g/mL; intervention of 15.6, 31.2 and 62.5 滋 g/mL of ETPB for 48 hours ahead of oxidative damage model preparation significantly increased the viability and SOD activity and decreased the MDA concentration in INS-1 cells (P〈0.05 or P〈0.01). Our results demonstrate the protective effect of ETPB against oxidative damage in INS-1 cells.
