无血清悬浮培养BHK21细胞增殖伪狂犬病毒的参数研究

Determination of Parameters of Pseudorabies Virus Proliferation with BHK21 Cells Cultured in Serum-free Suspension Condition

摘要: 旨在筛选适合生物反应器悬浮培养BHK21细胞的无血清培养基,并利用该技术培养伪狂犬病毒。通过筛选市售的无血清培养基,以细胞形态、细胞活率和增殖倍数作为驯化指标,对BHK21细胞进行无血清驯化;对使用10 L和40 L生物反应器无血清悬浮培养BHK21细胞的参数进行研究;同时利用筛选的无血清培养基和悬浮细胞进行伪狂犬病毒增殖测试。结果表明,市售培养基Ⅰ符合筛选要求,其培养的细胞形态比较均一,结团少,细胞生长2 d的增殖倍数为3.5倍,细胞活率在95%以上;生物反应器悬浮培养BHK21细胞的最适参数为转速80 r/min、pH值7.2、DO 60%;利用该工艺技术培养伪狂犬病毒,当接种细胞密度为6×106 cells/mL,接毒剂量为0.3%时,48 h收获的病毒液滴度可达109.5 TCID50/mL。该试验实现了伪狂犬病毒的无血清悬浮生产,为其生产工艺的优化提供了可能。

Abstract: The objectives of this study were to screen a serum-free medium suitable for suspension culture of BHK21 cells in bioreactor, and to achieve pseudorabies virus proliferation by using this this technique. BHK21 cells were serum-freely acclimated by screening the commercially available serum-free medium using cell morphology, viability and multiplication ratio as evaluating indicators. The parameters of serum-free suspension culture of BHK21 cells were determined in 10 L and 40 L bioreactors. Furthermore, a pseudorabies virus proliferation test was carried out by using the screened serum-free culture medium and suspending cultured BHK21 cells. The results showed that the commercially available mediumⅠwas consistent with the screening requirements, and the cultured cells were uniform in morphology and less in clusters. The proliferation ratio of the cells cultured for 2 days was 3.5 times, and the cell viability was above 95%. The optimum parameters of suspension culture of BHK21 cells in bioreactor were as follows the rotation speed was 80 r/min, pH value was 7.2, and DO was 60%. Under this condition, when the inoculated BHK21 cell density was 6×106 cells/mL and the inoculated pseudorabies virus dose was 0.3%, the titer of the harvested viruses at 48 h reached up to 109.5 TCID50/mL. The serum-free suspension proliferation technique of pseudorabies virus was achieved in this study, providing the possibility of optimizing its production process.
