  • Study on Isolation and Drug Resistance of Staphylococcus aureus from Dairy Farm

    In order to understand distribution and drug resistance of Staphylococcus aureus of Henan Province. 400 samples were collected from dairy farms, and S. aureus were isolated and identified by VITEK-2 Compact automatic microbial analysis system,and the drug resistance analysis was carried out by mini-broth dilution susceptibility test. The result showed that total 82 isolates of S. aureus were isolated, and the isolating rate was 20.5%. The drug resistance analysis results showed that S. aureus had serious resistance to penicillin G, erythromycin and ampicillin, and the drug resistance rates were 100%, 86.6% and 98.8%. S. aureus was sensitive to cephalosporin, aminoglycoside and fluoroquinolone antibiotics.It is indicated that animal-origin S. aureus of Henan Province had serious resistance to antibiotics, so antibiotics should be cautious in animal production.
    Efficacy Test on Ligularia virgaurea in Grassland with Two Kinds of Herbicide

    Two kinds of herbicide were used to control Ligularia virgaurea in grassland. The results showed that the control rate of Milestone at the dose of 500.0 mL/hm2 on L. virgaurea in grassland was more than 80%, which was significantly higher than that with other two doses of Milestone and Bentazone. The control effect of Bentazone was worse and it was safe to most Gramineae and Cyperaceae grass. Milestone at the dose of 500.0 mL/hm2 could be used and popularized as an effective medicament against L.virgaurea.
    Study on the Estimation Model of Leaf Biomass of Single-plant Buxus microphylla

    The crown breadth, plant height and leaf biomass of 44 plants of Buxus microphylla were measured in the test. 34 plants were used to establish the model among them. The binary regression analysis of leaf biomass was made by using Excel 2003 to establish the estimate formula of leaf biomass of B. microphylla. And other nine plants were used to validate the model. The results showed that the leaf biomass of B. microphylla had no significant correlation with the plant height(P0.05). The leaf biomass showed a significant correlation with the crown breadth(P0.05). The estimation formula of leaf biomass of B. microphylla was as follows: Y(g, leaf biomass)=156.12+7.332X(cm, crown breadth,P0.05).

    [Objective] The research aimed to optimize the optimum water-extracting technology of allergic rhinitis granule.[Method] Taking the yield of dry extract and paeoniflorin content as evaluation indices, the extraction technology of allergic rhinitis granule was studied by using L9(3^4)orthogonal experiment. [Result] The optimum extraction technology conditions were as follows:adding water with the amount 8 times as the amount of medicinal materials, decocting for 2.0 h for 3 times.[Conclusion] Water-extracting technology was simple,scientific, reasonable and feasible. The production cost could be reduced by using water-extracting technology. And the water-extracting technology could be applied in industrial production on the basis of ensuring the stable clinical curative effect of allergic rhinitis granule.

    In order to ensure the comparability and reliability of animal test results, the stability of quality of tested animal must be kept, so it is very necessary to detect the quality of tested animals. One mouse was taken from 13 separation facilities of Compare Medical Center of Yangzhou University separately, and contents of ileocecal junction and life waste were collected and inoculated into responsible mediums. According to GB/T 14926.14 and GB/T 14926.17-2001, Staphylococcus aureus and Pseudomonas aeruginosa were detected. Results showed that the Staphylococcus aureus was positive in some mice, while Pseudomonas aeruginosa was negative in all mice. It is suggested that a part of mice in separation facilities of Compare Medical Center of Yangzhou University took Staphylococcus aureus, but there was no Pseudomonas aeruginosa.

    In order to investigate the effects of goat placental transfer factor injection(GPTF) on non-special immunological regulating function, chicken RBC phagocytosis test, carbon clearance test and release of immune medium by macrophage induced with GPTF were carried out. The results showed that GPTF could advance the phagocytosis of macrophage to engulf chicken RBC, improve the indexes of clearance and phagocytosis of immune system of tested mice, and increase macrophage secretary of NO and IL-1. It is indicated that GPTF possesses a role to regulate the non-specific immunity in animals.
    Determination Method Establishment of Polysaccharide Content in Acanthopanax senticosus

    The research aimed to determine the polysaccharide content in medicinal plant Acanthopanax senticosus and provide scientific basis for the quality control and resource development of A. senticosus. Taking glucose as control, the polysaccharide content in A. senticosus was determined by using phenol-sulfuric acid method. The results showed that the standard curve of glucose was A=0.0133C-0.0025(R2=0.997 5). The polysaccharide content was 2.40%. The method had good precision, and the adsorption of reaction products was stable within 2 hours, and the average recovery was 99.1%.
    Determination of the Phylogenetic Relationship of Equus Animals Based on Three Mitochondrial Genes

    [Objective] Because abundant fossil evidences of Equus animals were discovered, it has become the best example of species evolution. However, there are still other troublesome issues in fossil evidences,such as the gaps in the fossil record and difficulty to distinguish from each other. The aim of this study was to provide some understandings for the genetic differentiation and genetic relationship identification of Equus animals at molecular level, provide some helps for the protection, exploitation and utilization of Equus animals and provide basic teaching materials for the teaching of biology. [Method] The molecular biology softwares were used to align the mitochondrial DNA(mtDNA)sequences in GenBank. Single nucleotide polymorphism(SNP)sites were detected. And the phylogenetic analysis based on SNPs was conducted. [Result] Abundant genetic variations were found in three mitochondrial genes(COX1, Cytb and ND4), which could reveal the genetic relationship of Equus animals.[Conclusion] The topological structures of the constructed phylogenetic trees based on the three genes were basically the same. In the three trees, white rhinoceros sequences were all placed as the outgroup. And Przewalskii horse sequence was clustered with horse sequence firstly. Then the three kind of Asian wild ass were clustered. All zebra sequences were clustered firstly and then they were clustered with the donkey sequence.

    Yanchi Office of Science and Technology introduced planting techniques of Astragalus membranaceus from 2011 to2013, carried out a planting experiment of A. membranaceus in Yanghuang irrigation area and Sixteen Fort dry sand of Yanchi arid and sandstorm area, and got good economic benefit. Meanwhile some cultivation techniques of A. membranaceus were summarized combining with local climate and soil conditions in order to popular and apply A. membranaceus in similar areas.
    Effects of Grazing Pattern on the Anatomical Structure Characteristics of Cleistengenes songorica Leaves

    The leaf anatomical structure of Cleistengenes songorica under different grazing patterns were studied. The results showed that the skin in the leaf of C. songorica had a layer of cells. Mesophyll cells were short-column, and epidermal cells were vertically arranged. The midrib thickness, blade thickness and mesophyll cell area of C. songorica in night-grazing region and free-grazing region were lower than that in day-grazing region, and the area of epidermal cells and thickness of corneous layer were higher than that in day-grazing region.

    miRNA, with full name of microRNA, is a kind of conservative non-coding single-chain RNA molecular, and its length was about 22 nucleotides(nt). miRNA come from pre-miRNA of about 70 nt through Dicer enzyme cutting. miRNA is small, but it could combine with target mRNA and then form into complete or incomplete complementary condition to lead silencing complex to degrade mRNA or forbid its translation so as to play gene regulatory effect. miRNA plays an important role in animal growth,hormones secretion, cellular differentiation, cell proliferation and death, and carcinogenesis. The function of miRNA and correlation researches between miRNA and occurrence, development and transfering of tumour were reviewed.

    The transcription factor, carbohydrate response element binding protein(ChREBP), is combined with several relevant genes of glycolysis and lipogenesis to regulate glucose and lipid metabolism. And glucose and lipid metabolism is the focus on metabolic syndrome such as diabetes and obesity for research, and it is also the concerns of improving the quality of meat,cultivating high quality of livestock and poultry breed. The structure of ChREBP, tissue distribution, biological function and influencing factors on its expression were reviewed in order to provide theoretical basis for regulating glucose and lipid metabolism better.

    Methane produced by rumen fermentation of ruminants was excreted from mouth by eructation, which reduced the feed utilization rate and aggravated the greenhouse effect. Therefore, it was of important significance for the economy and environment to reduce the yield of methane produced from the rumen of ruminants. The excretion amount of methane from ruminants, the influencing factors of the excretion amount of methane and the kinds of methane inhibitors were mainly reviewed.

    The reasons for fail artificial propagation of Carrulax conorus were analyzed:the food nutrition imbalance may be the primary problem to affect artificial propagation of C. conorus, and the change of living environment may be main reason for influencing artificial propagation of C. conorus under human feeding condition, such as light, temperature, humidity, the scope of activities, mating conditions. Based on the analysis of choosing and breeding of C. conorus, constructing the house and nest of C.conorus and food filling, it is concluded that the artificial propagation of C. conorus is feasible only if some conditions of artificial feeding were satisfied, such as qualified dear bird, suitable environment, appropriate nest and reasonable diet.

    HACCP is a preventive control management system. Each production link in processing of chilled pork were analyzed by using the principle of HACCP. Five critical control points(CCPs) were confirmed as checking of live pigs, slaughtering and segmenting, inspecting after slaughter, metal detecting, cooling and discharging acid. And some corresponding control and correction measures were established. And an effective HACCP scheme was confirmed and built up finally. The application of HACCP system in the production process of chilled pork supplied for Hong Kong and Macau was mainly discussed, so as to better control the production of chilled pork supplied for Hong Kong and Macau and guarantee the food security by applying HACCP system.

    60 yak calves with similar month-age were selected to make full-lactation and fattening tests. The effects of full-lactation on the growth and development of yak calves were studied. The results showed that full- lactation could not only increase the body weight of calves, but it could also improve the economic benefits of farmers.

    [Objective] This experiment was conducted to investigate the effects of small-enclosure rotation feeding pattern on the welfare of beef cattle. [Method] 12 healthy and castrated Limousin crossbred cattle with similar weight of(420.6±23.4) kg were randomly divided into two groups. Small enclosure rotation feeding pattern was used in group A and traditional feeding pattern was used in group B. The frequency of lying behaviour, lying time, ruminating time and other indices of beef cattle were determined.[Result] The lying frequency lying behaviour, lying time and ruminating time of beef cattle in group A were significantly or extremely significantly higher than those in group B(P0.05 or P0.01). [Conclusion] Under these conditions, the lying time and ruminating time could be significantly improved by small-enclosure rotation feeding pattern, so as to improve the welfare of beef cattle.

    In order to detect and investigate the morbidity of mastitis of lactating cows in a dairy farm in Korla City of Xinjiang, the subclinical mastitis of 32 lactating cows in this dairy farm was detected. The results showed that the positive rate of bovine subclinical mastitis was 40.63%, and the positive rate of mastitis increased with the increasing of the reproductive cycles of the cow. The positive rate of mastitis in dairy cows with milking machine was higher than that with manual milking. And the positive rate of mastitis at back mammary gland was higher than that at front mammary gland.
