  • Determination of the Antibacterial Activity of Extract of Forsythia suspensa to Common Pathogenic Bacteria in vitro and in vivo

    In order to observe the antibacterial activity of Forsythia suspense extract on Staphylococcus aureus, Escherichia coli, Salmonella sp. and other pathogens, the antibacterial activities of F. suspensa extract on S. aureus, E. coli and Salmonella sp. in vitro were determined by using tube double-dilution method. After mice were administrated with the same concentrations of pathogens by intragastric administration, mice were treated with F. suspensa extract to determine the number of bacteria in the duodenum, jejunum, ileum and cecum of mice. The results showed that F. suspensa extract had inhibitory effects on S. aureus, E. coli and Salmonella sp. in vitro and in vivo. And the inhibitory effect on S. aureus was the strongest and that on Salmonella sp. was the weakest. The mortality of mice induced by pathogens was significantly reduced by perfusing F. suspensa extract. In conclusion, F. suspensa extract had obvious inhibitory effects on S. aureus, E. coli, Salmonella sp. and other common pathogens in vivo and in vitro.
    Evaluation and Comparison of Water Quality of Sanya Bay

    Water quality was affected by the quick development of coastal tourism and aquaculture of Sanya City in recent years. Some water quality parameters of pH value, dissolved oxygen, phosphate, nitrite, total phosphorus and chemical oxygen demand were measured in coastal water of Sanya Bay, close to urban district, and Yalong Bay, far from urban district, from January to April in 2013 for assessment of water quality of coastal water of Sanya City. Concentrations of phosphate, nitrite, total phosphorus and chemical oxygen demand of coastal water of Sanya Bay were higher than that of Yalong Bay, and there were significant difference (P〈0.05) and very significant difference (P〈0.01) in some months. Value of pH and dissolved oxygen of both bays indicated that water quality of both bays were consistent withⅠ-Ⅱkind sea water quality standards. Phosphate concentration of Sanya Bay and Yalong Bay were 0.015 2-0.018 9 mg/L and 0.008 2-0.014 3 mg/L respectively, which were consistent withⅡ-Ⅲkind andⅠsea water quality standards, respectively. Chemical oxygen demand of Sanya Bay and Yalong Bay were 0.528-0.899 mg/L and 0.533-1.013 mg/L respectively, and both were consistent with Ⅰsea water quality standards. Water quality of coastal water of Yalong Bay was better than that of Sanya Bay, and water quality of former was consistent withⅠkind sea water quality standard in most coastal water area, and the latter was consistent withⅡ-Ⅲkind andⅠkind sea water quality standard in most coastal water area and part coastal water area respectively, in tourism season.

    The research aimed to study the effects of different temperature and reaction time on the antigen titer of H5N1 subtype of avian influenza Re-6 strain determined by hemagglutination test. Three temperatures (4, 25 and 37℃) were set to read the titers after reacting 5, 10, 15, 20, 25, 30, 35, 40, 45, 50, 55 and 60 min respectively. The results showed that the the appearance time of result was shortened with the increase of reaction temperature. The results tended to be stable after reacting at 4℃for 25 min and deflocculation started after reacting 50 min. The results tended to be stable after reacting at 25 ℃ for 20 min and deflocculation started after reacting 40 min. The results tended to be stable after reacting at 37 ℃for 15 min and deflocculation started after reacting 30 min.

    Sixty hybrid lambs of Mongolia sheep and Dorper sheep (Dumont hybrid lambs) which had similar birth weight and birth date and normal growth and development were chosen, and effect of early weaning of Dumont hybrid lambs on growth and development was studied. The results showed that from birth to 4-age period, the weight absolute growth and body size (body height, body length, chest girth and cannon circumference)of Dumont hybrid lambs increased quickly, so the period was peak of growth for Dumont hybrid lambs. According to the growth and development rule of Dumont hybrid lambs, it has important significance in development of local animal husbandry to strengthen the nutritional requirements and feeding management of lamb growth peak.
    Effects of Different Concentrations of G418 on Ear Fibroblast Cells of Sheep

    In order to find the lowest concentration of G418 to kill ear fibroblast cells of sheep from 10 days to 14 days, effects of G418 of 10 concentrations at 100-1 000 μg/mL on ear fibroblast cells of sheep were studied. Results showed that the G418 at 200-400μg/mL could kill all ear fibroblast cells of sheep from 10 days to 14 days.
    Histological Observation on the Liver and Kidney of Achatina fulica

    The organizational structure of the liver and kidney of Achatina fulica was observed with microscopy by using conventional slicing technology. The results showed that the liver surface of A. fulica was covered with fibrous connectivetissue. And its outside was serosa and there were more visible adipose tissue in the submucosa. Connective tissues existed among hepatic lobules. And hepatic vein was observed and obvious liver cell cord wasn′t found. The fiber membrane coated on the kidney surface was close to the cortex and smooth muscle was in the underneath. And visible kidney tubules were seen and the lumen of kidney tubules was narrow.
    Study on the Effects of Different Salt Concentration, Culture Media and Trace Elements on the Degradation of Paraffin Wax by Paraffin-thirsty Bacteria PWⅠ

    The degradation effects of paraffin by paraffin-thirsty bacteria PWⅠ under the concentrations of different salt concentration, culture media and trace elements were studied. The results showed that the degradation rate of paraffin by paraffin-thirsty bacteria PWⅠreached the maximum (47.31%) when salt concentration was 1%. And the degradation rate of paraffin in liquid culture medium of paraffin was 47.76%. Different trace elements had obvious effects on the degradation of paraffin. The degradation rates of paraffin when Cu2+concentration, Zn2+concentration and Fe2+concentration were 0.10, 0.07 and 0.55μmol/L respectively reached the maximum values, being 48.30%, 47.95% and 45.26% respectively. These results will be helpful for applying PWⅠin paraffin removal and control of oil well in biological technology and enhancing the oil productivity of oil well.
    Study on the Isolation, Identification and Characteristics of Rhodotorula mucilaginosa in Alhagi sparsifolia Silage

    Alhagi sparsifolia silage was diluted and coated to obtain single bacteria. And single bacteria were purified on PDA medium to obtain three stains of Gram positive bacteria. And the colony morphological identification, physiological and biological properties and ITS (ITS1+5.8S+ITS2)sequencing homology analysis were made on the positive bacteria. The results showed that one strain of red single colony was round or oval,and its centre was ridgy. This stain was able to utilize varied of carbon sources (sucrose, D-galactose, etc.) and two kinds of nitrogen sources [(NH4)2SO4 and L-glutamic acid]. But this strain wasn′t able to utilize KNO3. The whole sequence length of 5.8S rDNA was 616 bp. The homology between this strain and Rhodotorula mucilaginosa ATCC 4054(NCBI accession number:KC881069)was 99.8%by DNA Star Software. So this strain was identified as Rhodotorula mucilaginosa.

    Objective] The research aimed to detect cattle and sheep-derived components in the feed of ruminants and animal-derived feed. [Method]The cattle and sheep-derived components in 131 batches of the feed products (premix, concentrate supplement, complete feed, etc.) of ruminants and animal-derived feed (fish meal,poultry meat powder, pork powder, feather meal, etc.) were detected by real-time fluorescence qualitative PCR. [Result] No cattle and sheep-derived component was detected in all samples. [Conclusion] No cattle and sheep-derived component was found in the detected feed products of ruminants and animal-derived feed. The monitoring of cattle and sheep-derived components should still be strengthened.
    Contrast Study on Different Compound Feed on Production Performance of Fattening Pig

    Objective] The purpose was to screen out feed varieties suitable for test pig farm production through comparing feeding and benefit of two different kinds of compound feed. [Method] Forty pigs with weight of (47.3±2.5) kg were averagely divided into two groups and fed with two different kinds of compound feed for 55 days respectively. When the weight of fattening pigs reached about 90 kg, their production performances were analyzed. [Result] After the experiment was finished, each piglet average final weight and each piglet average total weight gain of Dabeinong (DBN)feed group of fattening pigs were 11.29%and 16.32%higher than that of Jingeli(JGL)feed group respectively(P〈0.01), but the ratio of feed and meat of DBN feed group was 13.64%lower than that of JGL feed group (P〈0.01). [Conclusion] These results indicated that the compound feed of DBN were more suitable for feeding fattening pigs from the above factors and economic benefits of the pig farms.

    Probiotics are active preparation, composed of living microorganisms. Researches showed that it has a variety of useful characters to animal health, such as good effect,no residues etc. Thus, it has become a research hotspot around the world, and many different products have been developed on the market. Probiotics show different biological activity, and play important roles in preventing disease, nutrition regulation, immune regulatory whether used alone or mixed on animal body. Therefore, it will become an inevitable trend that probiotics replace antibiotics in breeding livestock and poultry. The definition, origin and development of probiotics and progress on anti-disease nutrition of livestock and poultry breeding in recent years were reviewed.

    Taking composite polymer material as coating wall and multivitamins particles of lactation cows as core materials, rumen-protected multivitamin were prepared through granulating and coating with composite polymer by fluidized bed. Results showed that the optimal coating and core proportions of RP-multivitamin product according to the technology parameters were 6%-8%.

    The uncoupling proteins 2 (UCP2) is one kind of protein which has uncoupling function. The general belief is that UCP2 is related to the fatty acid metabolism and ATP generation. The expression of UCP2 gene in different visceral tissues of small-tailed Han sheep at three age stages (6,8 and 10 months of age) was analyzed by real-time quantitative PCR technique. The result showed that ages did not affect the expression of UCP2 gene in the same visceral tissue (P〉0.05);the expression level of UCP2 gene in spleen was significantly higher than that in heart, kidney and rumen (P〈0.05), but there was no significant difference in heart, kidney and rumen (P〉0.05). This experimental results support the fact of wide expression of UCP2 gene in visceral tissue of small-tailed Han sheep and the viewpoint that there is an association between UCP2 gene and the immunity. Furthermore, it is indicated that ages has little effect on the expression of UCP2 gene in the visceral tissue of small-tailed Han sheep at 6-10 months of age.

    The control effects of Milestone on Oxytropis ochrocephala Bunge were tested. The results showed that obvious control effects were obtained in two treatments of Milestone on O. ochrocephala after pesticide application 30 and 60 days and the efficacy were both more than 70%. The efficacy on O. ochrocephala with 500 mL/hm2 Milestone was over 80%. Two concentrations of Milestone was safe to pasture, so they could be used and popularized as effective control pesticide to O. ochrocephala.

    The effects of replacing part of meat rabbit′s feed with Italian ryegrass and wild grass on the weight gain, slaughter traits and meat quality of rabbits were studied. It was found after feeding 40 days that replacing 10%-50%concentrate feed with Italian ryegrass or wild grass had no significant effect on the weight gain of meat rabbits(P〉0.05). The application of greenfeed improved the slaughter rate of meat rabbits, obviously reduced the fat content in rabbit meat and improved the quality of rabbit meat.

    According to objectives of university personnel training, combined with the psychological characteristics of undergraduate study, the teaching content and methods of young university teachers teaching Animal Anatomy course were analyzed.
