  • Study on Rapid Identification of Milk Protein by Polyacrylamide Gel Electrophoresis

    The SDS-polyacrylamide gel electrophoresis was taken to the rapid detection research on milk quality. Through the exploration on conditions such as dilution multiple, voltage conditions, dyeing time, washing time and times, and the milk protein electrophore-togram with clear bands was obtained. The test result showed that, the optimum condition of milk protein identification with rapid polyacrylamide gel electrophoresis was that 8 % separation gel was used for electrophoresis under 120 V and the protein electrophoresis dyeing solution with super sensitivity was taken to rapidly stain, with boiling for 1 min and shaking for 10 min, which could realize the aim of rapid identification of milk protein.
    Correlation Analysis of lnterspecific Relationship of Community in S@a breviflora Desert Steppe in Siziwang Banner in Inner Mongolia

    In order to explore the interspecific relationship of the community in Stipa breviflora desert steppe in Siziwang Banner, Inner Mongolia, on the base of sample plots investigation on its community, the interspecific correlations of 105 species pairs from 15 kinds of plants were analyzed by using research methods of X^2 test, Pearson coefficient and Spearman rank correlation coefficient. The result showed that, the most interspecific community in Stipa breviflora desert steppe in Siziwang Banner had the negative correlation. The number of species pairs with significant and extremely significant correlation was little. The most of species pairs between dominant species showed significantly negative correlation and the number of species pairs between the dominant species and other species with negative correlation was more. It indicated that species niche of the community was differentiated and the species was independent, but the competition to resource availability was fierce and the community was in development and succession.
    Study on Optimum Culture Time for the Everted Gut Sacs in Broilers

    The test was to explore the optimum culture time for the everted gut sacs in broilers by the lactate dehydrogenase method. Using single factor test design, the duodenal, jejunum, ileum, intestine of 6 Three-yellow cocks were distributed randomly into conical flask with a broiler as a repeat and a intestinal segments as 6 repeats. Then at 0, 20, 40, 60 and 80 min, 100 uL culture solution in each conical flask was taken with a pipette gun to analyze the activity of lactate dehydrogenase. The result showed that, the activity of lactate dehydrogenase in culture solution was increased slowly from 0 min to 60 min and the rising range was faster after 60 min. The optimum culture time for the everted gut sacs in broilers was 40-50 min.

    With the Stipa breviflora desert steppe in Siziwang Banner of Inner Mongolia as the research object, the pattern distribution types, pattern scale and clump intensity of 3 kinds of dominant plants as S. breviflora, Cleistogenes mutica, A rtemisiafrigida with different stocking rates were studied by using t test of diffusion coefficient, mean crowding intensity, clumping index, patchiness index, Green index, Cassie index and so on. The results showed that the distribution types of S. breviflora, C. mutica, A. frigida were clump distribution. In CK area and middle pasturing area the clump intensity of 3 kinds of plant in order was C. mutica 〉 A. frigida 〉 S. breviflora and in light and heavy pasturing area, their clump intensity in order was A. frigida 〉 C. mutica 〉 S. breviflora. With the degradation of desert steppe, C. mutica and A. frigida would become as the main populations in community.

    On the base of data investigation with point-centred quarter method, the community structure composition of Tamarix elongata distributed on sale dune in middle reaches of Shule River was described. The association relationship of spatial pattern and interspecific ecological distribution of the dominant species in community was studied by using the methods of theoretical fitting by discreted distribution model, aggregate intensity indexes and interspecific association detection.

    The rise of temperature and the increase of precipitation were beneficial to increase the growth and development ability of forage. But the incongruity of the combination of the main climatic factors such as water and heat and the strong evaporation were not beneficial to the growth of forage, which hindered the growth and development of grassland plant in this region, decrease the primary productivity and increase the area of grassland degradation.

    The feasibility of establishment on HACCP system in healthy cultivation process of livestock and pouhry was studied. According the HACCP basic principle and application rules, the hazard analysis on each step of healthy cultivation of livestock and poultry was done and the HACCP system in healthy cultivation process of livestock and poultry was established. Using the method of critical control point judging-tree, 6 critical control points in the cultivation process such as introduction, feed, veterinary drug, vaccine, feeding management and sale loading were determined and the HACCP plan was made in detail.

    Brucellosis, caused by Brucella, was a infectious diseases of livestock and human chronic bacteria widely distributed in the world with the abortion and fever as characteristics and seriously threated to the life healthy of human and many kinds of animal. Brucella could infected human, many kinds of livestock and wild animals and resulted in enormous economic loss and serious public health problem. Thus, the testing work of Brucella was the important part to prevent its occurrence. The author summarized the detection technology of Brucella such as immunology ELISA, colloidal gold, PCR. Those technologies all could detect the Brucclla accurately, quickly, and specifically.

    In the test, the contents of Cu, Fe, Mn, Zn in the basal mixed diet of cow in dry period in the eastern of Hebei was determined by using atomic absorption spectrometry and its surplus and deficiency was analyzed to provide the reference for the formulation of cow diet in the dry period. The result of test showed that, the contents of Cu, Fe, Mn, Zn in the basal mixed diet were 299.87, 14.38, 37.70 and 32.67 mg/kg resp. The Fe content was 16 times of the Fe requirement of cow in dry period reported in NRC (2001) and 3 times of the requirement reported in the other literatures. The Cu content reached the requirement of cow in the dry period. The Mn, Zn content reached the requirement of cow in the dry period reported in NRC (2001) and was lower than the recommended requirement in the literature.

    1728 mammary regions from 451 production cows reared in a cattle farm of Baotou City in Inner Mongolia were detected, and the isolation identification and drug sensitivity test of pathogen were made in the experiment. The result showed that there are 252 cows suffered from recessive mastitis and the positive rate was 55.9 %; there were 1728 effective mammary regions, 480 positive mammary region and the positive rate of mammary region was 27.8 %. Staphylococcus aureus, streptococcus, Escherichia coli, corynebacterium, salmonella and Staphylococcus epidermidis were isolated from the milk of detected positive mammary region of rec, essive mastitis, resp. The detection rate of infectious bacteria of S. aureus and Streptococcus in the detected bacteria were higher. From the result of drug sensitivity test, cefazolin and quinolone drug had high sensitive to other bacteria except salmonella, and the common drugs showed tolerance to a different degree; the euphorbia Chinese herbal medicine had certain sensitivity to streptococcus, poor antibacterial effect on E. coli and staphylococcus, no antibacterial effect on corynebacterium and salmonella; Ruyanjinzhen was the mixture of Chinese traditional and Western medicine, which had better antibac, terial effect on the 6 pathogenic bacteria.
    Study on Ruminal Degradation and Intestinal Digestion of Different Carbohydrate Components from Maize and Soybean Meal by CNCPS

    The aim of the experiment was to study the ruminal degradation and intestinal digestion of different carbohydrate components from maize and soybean meal. The carbohydrate components in the entire samples of maize and soybean meal samples, ruminal undegraded residues and residues from digested small intestinal fluid freeze dried powder were subdivided by ruminal nylon bag technique and small intestinal fluid freeze dried powder digestion experiment combined with the CNCPS subdivision theory, and the characteristics of ruminal degradation and intestinal digestion of different components were discussed. The result showed that the rumen degradability of the dry matter from maize and soybean meal were 59.1% and 7.2.7 %, resp, and that of carbohydrate were 69.6 % and 66.2 %, resp. The small intestinal digestibility of the dry matter from ruminal undegraded residues were 40.8 %and 68.0 %, resp, and that of carbohydrate were 40.4 % and 62.7 %, resp. The ruminal nylon bag and small intestinal fluid freeze dried powder could be used as the method for studying the ruminant degradation, and combined with CNCPS for the component subdivision, it could be used to study the utilization of carbohydrate and its components better.
