Abstract: The regeneration system of potato variety Longshu No.5 was studied with stems as explants. The results showed that the optimal medium for callus induction from stem was MS＋2,4-D1.0mg/L＋NAA1.0mg/L respectively, the callus induction rates reached to 67.5% .The optimal medium for adventitious bud differentiation from stem segment-derived calluses was MS ＋ NAA0.5mg/L＋KT2.0mg/L＋GA30.Smg/L respectively, The adventitious bud differentiation rate reached to 66% .The optimal medium for rooting induction from tube plantlets was MS＋IBA0.5mg/L, and the rooting rate was 100%.
