Effect of Dimethyl Sulfoxide on The Activity of Tyrosinase

摘要: 在30℃、pH6.8Na2HPO4-NaH2PO4缓冲体系中,采用酶动力学方法研究了二甲基亚砜(DMS0)对酪氨酸酶二酚酶活力的作用。试验结果表明,当DMSO溶液体积分数≤25%时,随着DMSO溶液体积分数上升时,酪氨酸酶(TYR)活性相应上升;而当DMSO溶液体积分数〉25%后,TYR活性急剧降低。笔者探讨了DMSO对酪氨酸酶二酚酶活性作用机制,为TYR异常表达产生疾病的药物研发提供试验依据。

Abstract: The effect of dimethyl sulfoxide on the activity of mushroom tyrosinase was carried out by enzymological kinetic method with the condition of 25℃ and pH 6.8 phosphate solution buffer. The results showed that the diphenolase activity increased with increase of of DMSO based on the prior volume fraction was less than 25 % in the reaction system. Contrarily, the activity of diphenolase sharply decreased. The action mechanism of DMSO was explored in this paper, which provided scientific basis for development of medicines to treat the diseases associated with tyrosinase-induced.
