地锦粗提物对脾虚小鼠抗氧化能力影响的试验

Effect of Crude Extracts from Herba Euphorbiae humifusae on Antioxidant Index of Spleen-deficiency of Mice

摘要: 为了探讨地锦粗提物对脾虚小鼠脏器抗氧化能力的影响。采用泻下加劳倦过度的方法建立脾虚模型,造模时间为60d,期间测定谷胱甘肽过氧化物酶(GSH-PX)和血浆淀粉酶活性,二者活性显著降低后为脾虚模型建立。建立脾虚模型前和脾虚模型建立后相比较:脾虚组和地锦组小鼠的血浆淀粉酶活力均降低,均差异显著(P〈0.05);脾虚组和地锦组的血浆GSH-PX活力均降低,差异显著(P〈0.05)或极显著(P〈0.01)。将脾虚模型小鼠随机分为脾虚组和地锦组,同时设对照组。地锦组每天给予地锦水煎剂30g/(kg·bw),脾虚组和对照组给予相同剂量的生理盐水,试验期为60d。试验结束后,剖杀各组小鼠,取内脏,测超氧化物歧化酶(SOD)的活性和丙二醛(MDA)的含量。结果表明,脾虚组和地锦组小鼠各脏器SOD的活力均低于对照组,差异显著(P〈0.05)或极显著(P〈0.01),且随地锦粗提物试验期的增加,地锦组小鼠SOD的活力较脾虚组高,差异显著(P〈0.05);脾虚组和地锦组MDA的含量较对照组高,且差异显著(P〈0.05)或极显著(P〈0.01),随地锦粗提物试验期的增加,脾虚组MDA的含量较地锦组高,差异显著(P〈0.05)。表明地锦水煎剂粗提物可明显提高脾虚症状小鼠抗氧化能力。

Abstract: This experiment was designed to study the effect of crude extracts from Herba E.humifusae on antioxidant index of mice with spleen-deficiency.The modle was set up to produce splenasthenic syndrome by adopts lapactic and exhaustion for 60 days,the established modle was ensured by determining the activity of GSH-PX and amylase in plasma,when they significantly decline(P〈0.05)or(P〈0.01)compared with that of the former,then mice which is spleen-deficiency were randomly divided into the group of spleen-deficiency(SD) and the group of E.humifusae(EH),and the control group(C) also was settled.The animals of EH was given decoction of 30 g/(kg·bw),while the same dosage of sodium chloride in SD and C.60 days later,the trial animals were treated,the content of SOD and MDA was detected.The result showed that SOD activity in SD and EH is markedly lower than that of C(P〈0.05),with the elongation of using crude extracts from Herba E.humifusae,SOD activity was markedly higher than that of SD(P〈0.05);Meanwhile,MDA activity in SD and EH is markedly higher than that of C(P〈0.05),with the elongation of using crude extracts from Herba E.humifusae,MDA activity was markedly higher than that of EH(P〈0.05).The conclusion could be made that the crude extracts of Herba E.humifusae can enhanced the anti-oxidation activity obviously.
