Comparison on AFB1 Content in Feed Determined by ELISA and TLC Methods

Abstract: [Objective] The aim was to study the characteristics of ELISA method and thin layer chromatography(TLC) and compare advantages and disadvantages of aflatoxin B1 content determined by the two ways.[Method] The content of aflatoxin B1 was determined by using aflatoxin B1 enzyme league immune in ELISA method,while it was determined by using thin fluorescence scanning method in TLC method.[Result] The standard curve r value of aflatoxin B1 content determined by ELISA method was 0.999 3-0.999 9;the variation coefficient was 0.3%-0.6%,and the recovery was between 94.0% and 105.0%.TLC was half quantitative chromatography,but the standard curve r value of aflatoxin B1 content determined by using fluorescence scanning method was 0.998 1,and the recovery was between 84.60% and 90.08%.[Conclusion] Compared the two methods,TLC is traditional classical method,has advantages of strong ability anti-interference dream,offline testing and repeatedly test,but it also has more troubles before treatment,and has lower precision and recovery than ELISA method;ELISA is the international advanced method to detect AFB1,and has advantages of high sensitivity,less disturbance,simple,quick and safe operation.
