  • Effects of Different Separation Methods on the Purity of Testis Stromal Cells of Sheep

    In order to find a high-purity separation method of testis stromal cells of sheep, effects of three different separation methods on the purity of testis stromal cells of sheep were compared. Results showed that mechanical separation combining with cell screening filtering method not only was simple, but also had more than 90% of purity of stromal cells.
    Analysis on Water Quality in Coastal Area for Tourism Activities in Yalong Bay in Tourist Rush Season

    The coastal tourism in Sanya city has developed in recent years, and coastal waters environment was affected. Some water quality parameters of pit, dissolved oxygen, nitrite, phosphates, total phosphorus and chemical oxygen demand were measured for evaluating the water quality affected by different coastal tourism activities from January to April in 2013 in Yalong Bay. Results showed as followed: concentration range of dissolved oxygen was 6.3-7.4 mg/L and the comparison of that was control area〉swimming area〉living area〉speed-boat cruising area (P〉0.05).Concentration range of nitrite was 0.003 8-0.005 7 mL/L and comparison of that was speed-boat cruising area 〉swimming area 〉living area〉control area,furthermore ,there was significant difference for the concentration of nitrite between speed-boat area and controlling area(P〈0.05 ). Concentration ranges of phosphates and total phosphorus were 0.007 5-0.012 7 mL/L and 0.011 4-0.062 7 mL/L respectively, and the comparison of that was speed-boat cruising area 〉 living area 〉swimming area 〉control area,in addition ,concentration of both was higher significantly in speed-boat cruising area than that in controlling area and swimming area (P〈0.05).Concentration range of chemical oxygen demand was 0.48-1.28 mL/L and the comparison of that was living area〉speed-boat cruising area〉swimming area〉control area (P〉0.05).Conclusions were as followed: water quality of Yalong Bay was accordance with Ⅰ water quality standards overall in tourist rush season, but meets Ⅱ water quality standards in specific months because of the impact of different coastal tourist activities. Activities of speed-boat cruising and living area had more influence than other coastal tourist activities in coastal area of Yalong Bay in tourism rush season.
    Experiment on Content Determination of Adenosine and Cordycepin from Wild Aweto in Qiaojia County of Yunnan Province by HPLC

    The wild eordyceps has been a hot topic in recent years. Because of its growth in harsh environment, low natural parasitic rate and formation of the complex, Aweto can only grow in very few places. According to the relevant reports, Aweto is distributed in Yunnan, Siehuan, Tibet, Qinghai, Gansu and a few other places, so the number of Aweto is very limited, ean not meet the needs of people. A kind of Aweto was found in Qiaojia County of Yunnan Province recently, and it had wider distribution and a large number. There was no report on research and utilization of the wild Aweto. Contents of adenosine and cordycepin from the Aweto were determined by HPLC.

    Four hundreds and thirty-fine 240-day old Sutai pigs and 760 240-day old white Duroc×Erhualian F2 pigs were killed, and serum were collected to detect blood glucose and glycation serum protein (GSP); individuals' genotype was identified by using Illumina porcine 60 K SNP chip; association analysis between whole-genome SNP and two traits were carried out; QTL location to affect blood glucose and GSP of Sutai pigs and white Duroe×Erhualian F2 pigs at 240 days old were carried out. Results showed that five QTL affecting blood glucose and GSP was located, all at 5% level, among them there were two false positive, and their QTL located in different chromosomal regions, and explained 3.719% and 3.724% of the phenotypic variance.
    Identification and Karyotype Analysis of Offspring of F1 Hybrids to Two Kinds of Clover

    Hybrids F1 from distant hybridization of Trifolium ambiguum Bieb ev. Mengnong No. 1 and T. repens were highly sterile plants,which were just flowering instead of seeding over the past two years of the field planting. However, it started to fructification from the third year. In order to explore the seeding mechanism of F1 ,the technology of root tip chromosome tableting was used to identify the somatic chromosome of F1's seeds, meanwhile the condition of the tableting technology was explored and optimized. The results turned out that the somatic chromosome number in the seeds of F1 ranged from 40 to 48, and some cells of individuals emerged into chromosome doubling phenomenon and mixoploid. The chromosome number of somatic cells in F1 hybrids was 40,and 48 in T. ambiguum Bieb cv. Mengnong No.1. The somatic chromosome number of the 6-3% seeds was 44, which was initially identified as the resulting of backcross of F1 hybrids with T. ambiguum Bieb ev. Mengnong No.1 ,and its karyotype formula was 2n=30m+ 14sin, which belonged to the type of 2A.
    Preliminary Studies on CYP3A Sub-enzyme Activities of Bactrian Camel

    CYP3A sub-enzymes participate in biotransformation of 45%-60% of commonly used drugs, and play an important role in regulating the interaction of body and external environment and maintaining homeostasis. In order to study the activity of hepatic CYP3A sub-enzymes, the hepatic microsome was prepared by the Ca^2+ precipitation method, and the activities of CYP3A sub-enzyme was preliminarily studied by using biochemical methods. The result showed that the protein content of hepatic microsome was (1.541±0.264) mg/mL, the content of total CYP was (0.412±0.063 ) nmol/mg, and the activity of erythromycin-N- demethylase was (1.159±0.001) nmol/(mg· min ), respectively. The results laid a good foundation for further research of metabolic activity of CYP3A subfamily in vitro by using probe drug detection.

    Taking Helianthus tuberosus and milk as substrates, using sensory score as the index, the production technology of H. tuberosus yoghurt was studied by using single factor test and orthogonal test. The results showed that the optimum technology conditions were as follows:the addition amount of H. tuberosus juice of 8%, the addition amount of sucrose of 8%, the inoculation amount of starter of 3%, fermentation time of 4 h. The sensory score of the produced H. tuberosus yoghurt under the optimum conditions was 94.
    Effects of Milk Replacer on the Body Weight and Body Size of Early-weaned Calves

    24 Hostein dairy calves with the similar body weight were divided into two groups (12 calls in each group): test group and control group. The calves in test group were fed with beastings within five days after the birth, and then they were fed with milk replacer till the 42^nd day. The calves in control group were fed with milk before the 42^nd day. The results indicted that the average daily weight gain in test group was 836 g and that in control group was 679 g. The daily weight gain in test group was increased by 23.12% than that in control group.The increased values of chest girth, body height and body length of calves in test group were higher 2.90, 1.01 and 1.56 cm respectively than that in control group, without significant difference. The test group and the control group had no significant difference between the cannon circumference.It was indicated that milk replacer had good production effect, so it could take place of milk.
    Effects of Different Levels of Iron and Copper in Diet on the Growth and Development of Piglets

    The effects of different levels of iron and copper in diet on the growth and development of piglets were studied. One factor and four levels were used in iron treatments, and piglets were divided into one control group and four test groups (with four piglets in each group) and the test lasted for 28 days. Two-factor repeated test design was used in copper treatments and the piglets were randomly divided into four groups (20 piglets in each group) according to the sex and body weight, and the test lasted for 28 days. The results showed that the growth-promoting effects were better in 60 mg/kg yeast iron group. Adding 250 mg,/kg copper sulfate in the diet of weaned piglets could enhance the feed intake and weight gain of piglets and improve the feed utilization rate. Better economic benefits were obtained in the group of adding 5 mg/kg copper-lysine complex.

    The grassland resources in Naqu region of Tibet were lack of effective protection and scientific management and unilaterally pursued the number of livestocks in long-term traditional management mode of animal husbandry, which led to severe overgrazing phenomenon. The severe overgrazing phenomenon resulted in aggravated degradation of grassland resources, so it was difficult to maintain the sustainable development of grassland animal husbandry production. The existing problems in the current protection of grassland resources in Naqu region were discussed. And some suggestions were proposed for the protection and sustainable development of natural grassland in this region, so as to provide references for strengthening the grassland management, developing the ecological animal husbandry, achieving the harmonious development of grassland animal husbandry and the ecological environment of grassland.

    Artificial rearing and breeding is an effeetive method to protect wild animals (especially endangered animals). Though China has a long history of artificial rearing and breeding of wild animals, the rearing and breeding of wild animals has been developed for protecting resources and developing economy in recent years. Based on eontrasting the levels of planting forages and rearing and breeding wild animals at home and abroad, the contents, methods and achievements of constructing forage grass base of wild animals in Gansu Center for Proteetion of Endangered Animals of State Forestry Administration were summarized, so as to scientifically promote the development of grass industry teehnologies in local regions and their utilization in the protection of wild animals.
    Application of Microsatellite DNA in Paternity Identification of Yak

    To investigate the feasibility of microsatellite DNA in yak paternity identification applications, 17 microsatellite loci recommended by FAO were selected. The level of polymorphism of these microsatellite loci in Tianzhu White Yak groups was detected,and these 17 microsatellite loci for paternity exclusion probability cumulative was also calculated. Results were as followed: the average allele number of these microsatellite loci was 6.235 ;the polymorphism information content ranged from 0.191 to 0.824, with the average 0.608, showing high polymorphism;a candidate parent cumulative probability of exclusion was 0.997,and the two candidate parent cumulative probability of exclusion was 0.999 999 97;the null allele frequency of each microsatellite loci was below 0.05.Microsatellite DNA could be used for paternity identification in yak.

    Simmental is a dairy-beef dual-purpose breed and it is the second cattle breed in the world. Its present number in stock in the world is more than 41000 000. Inner Mongolia is the breeding base of Simmental in China. Nowadays Simmental has been the dominant species in beef cattle industry in Inner Mongolia. At the end of 2013, the number of beef cattle in stock in Inner Mongolia was over 5700 000, and more than 90% of fine breeds and improved breeds had Simmental bloodline. Based on analyzing the production characteristics of Simmental, some technology measures of rearing Simmental were proposed so as to provide references for breeding Simmental in Inner Mongolia.
    Fattening Trial of High Quality Black Goat

    In order to improve the benefit of raising goat by planting grass and the development of grassland ecological animal husbandry of Bijie City, a fattening trial of high quality Black Goat was carried out by using trace elements nutritive cube prepared by Bijie Institute of Animal and Veterinary Sciences of Guizhou Province. Result showed that the gaining effect of feeding concentrates combining with nutritive cube was obviously higher than the other two groups, and all the gaining rates were more than 50%.

    In order to understand the infection situations of Toxoplasma gondii of sheep and the relationship between infection of T. gondii and abortion of sheep in Aletai region, 565 serum samples were randomly collected from six counties and one city of Aletai. The infection situations of T. gondii were detected by using indirect hemagglutination test. The results showed that the infection of T. gondii of sheep occurred in each county (city) of Aletai. The infection rate was different and the average infection rate was 6.5%. The mixed infection ofBrucella melitensis and T. gondii was found in some sheep with the abortion history, which indicated that the infection of 7: gondii should be also considered in order to prevent and treat the abortion of sheep, in addition to the infection of B. melitensis.
