Callus Induction from Leaves and Stems of Semen Cassiae

Abstract: Semen Cassiae is an important medicinal plant in Mongolian medical practice.In this paper, influence of plant growth regulator (PGR) on callus induction from stems and leaves were determined by experiments. The optimized protocol was established as follows.The seeds of Semen Cassiae were disinfected with 75% alcohol for 45 S,and then 0.1% HgCl2 for 5 min; afterwards,they are placed onto the medium containing 30 g/L sucrose and 7 g/L agar.When we obtained the aseptic seedlings, the leaves were removed for callus induction and transferred onto medium containing of MS,30g/L sucrose,7g/L agar,2 mg/L 6- BA and 0.6 mg/L NAA;simultaneously,the stems were removed and transferred onto the medium containing MS, 30g/L sucrose, 7g/L agar,3mg/L 6-BA and 0.7 mg/L NAA.Our work will promote multiple propagation and cell culture of Semen Cassiae,since there has been no report about tissue culture of this medicinal plant in China
