槐花中芦丁的正交提取工艺及抗氧化活性研究

Study on Orthogonal Design Mode and Antioxidant Activity of Rutin from Sophora japonica

摘要: 以芦丁的紫外最大吸收波长510nm为指标,进行正交试验设计,筛选出从槐花中提取芦丁的最佳工艺参数,将芦丁用超氧负离子自由基清除法进行抗氧化活性试验。结果表明,醇提法提取工艺最为理想,其工艺参数为:乙醇浓度80%、乙醇用量100mL、振荡温度45℃,该工艺提取出的芦丁含量最高可达11.6%;芦丁对超氧负离子自由基O2-·的清除作用呈量效关系,清除作用的IC50值为:0.0305g/L,存在显著性差异(t=2.93,P〈0.05)。

Abstract: In the study,absorption maximum wavelength of 510 nm was used as index for the orthogonal design mode to screen the optimization parameter of rutin extraction from Sophora japonica,extracted rutin was used for super oxide anion radical method for antioxidant activity experiments.The results showed that alcohol extraction was the most ideal method,and its parameters was ethanol concentration of 80%,ethanol volume of 100 mL and ultrasound temperature of 45℃,rutin of 11.6% could be achieved by it.There was dose-dependant relation for rutin to clean up free radicle O2-·,and the scavenging index of IC50 was 0.030 5 g/L,which is significant differences exist(t=2.93,P0.05).
