脱水素基因转化紫花苜蓿及其初步抗旱性分析

Transformation of Dehydrin Gene into Alfalfa and Preliminarily Analysis of Its Drought Resistance

摘要: 为了获得抗旱性较强的转基因苜蓿,试验以紫花苜蓿品种“中苜2号”作为基因转化受体,将沙冬青脱水素基因通过农杆菌介导,转化到“中苜2号”中。试验结果表明,子叶和下胚轴作为外植体愈伤组织诱导频率最高,外植体与农杆菌的共培养时间以5d为最佳,试验共获得126株抗性再生植株;PCR和RT—PCR鉴定后,得到了11株阳性转基因苜蓿;通过干旱处理后,经初步的表型和脯氨酸分析显示,转基因苜蓿具有较强的抗旱能力。

Abstract: In order to obtain transgenic plants of alfalfa with stronger drought resistance ,dehydrin gene of A mmopiptanthus mongolicus was transformed into the genome of Zhongmu No. 2 mediated by Agrobacterium tumefaciens with alfalfa variety Zhongmu No. 2 as gene transformation receptor. The results showed that the induction frequency of cotyledon and hypocotyls as explant calli was the highest and the best co-culture time for explants and A. tumefaciens was 5 days. 126 resistant plants were obtained. After the identification by PCR and RT-PCR, 11 positive transgenic alfalfa plants were obtained. The elementary phenotype analysis and proline analysis after drought treatment indicated that transgenic alfalfa had stronger drought resistance ability.
