  • Evaluation of Soil Resource on Cultivated Land in Laixi Based on GIS

    Evaluation unit is formed by overlaping county soil data,present landuse map,agrotype map and administrative map;the appraisal factors were ensured including 13 factors from six aspects of production performance,land management,site condition,section composition,soil physical and chemical properties and the nutrient;the degree of membership is ensured by Terfel method and analytic hierarchy process by definite weight.The comprehensive evolution on farmland soil data of Laixi City was made with MAPGIS software.The result showed that the cultivated land fertility are mainly two,three and four,respectively accounting for the proportion of 28.0%,39.0%,20.7% in total cultivated area of Laixi City.

    In the study,small intestinal mucosal structure,muscular layer thickness and the distributions and numbers of intraepithelial lymphocytes(IELs) and goblet cells(GCs) from Sunit sheep at 6 months old age was determined by Histological and Histochemical methods.The results showed the length of Duodenum villus were the longest,its length attained(353.25±60.48) μm,the Jejunum and Ileum in villus length were(317.27±72.02) μm and(275.36±47.62) μm respectively,but there was markedly different between Jejunum and Ileum(P0.05);In mucosal thickness,Duodenum were the thickest reaching(669.15±139.04) μm,the Jejunum and Ileum were(586.72±134.50) μm and(551.85±91.32) μm,Duodenum in thickness was extremely different comapared with that of the Ileum and Jejunum(P0.01);In the thickness of muscular layer,Ileum was the thickest reaching(280.45±58.33) μm,Jejunum was(167.16±42.63) μm and Duodenum was(148.78±38.36) μm,the difference was marked among them(P0.01);In the depth of crypt,Duodenum was the deepest reaching(309.36±74.21) μm,Jejunum was(286.23±57.23) μm,the difference was marked among them(P0.01);In the value of V/C,Duodenum and Jejunum were(1.80±0.45) and(2.00±0.48) respectively,there was different between Jejunum and Ileum(P0.01);In the number of IELs and GCs,all parts of intestinal mucosal layer were not significantly different(P0.05).The conclusion could be made that the length of villus,the mucosal thickness and the depth of crypt were gradually become smaller,while the muscular layer thickness were gradually become greater in small intestine from anterior to posterior,but there were no change of IELs and GCs numbers in all parts of small intestine.

    A microwave digestion procedure was developed for trace Pb in cigarette adhesive using microwave digestion system,and the amounts of these trace metallic element was determined by atomic absorption spectrometry after microwave digestion.The recovery of Pb was on the top of 97% with the RSD of 0.5%.The results showed that this method was rapid,simple,minimized environmental contamination and the samples are completely digested,the chemical reagents were economized during the sample preparation.The precision and accuracy are satisfactory.

    In the study,absorption maximum wavelength of 510 nm was used as index for the orthogonal design mode to screen the optimization parameter of rutin extraction from Sophora japonica,extracted rutin was used for super oxide anion radical method for antioxidant activity experiments.The results showed that alcohol extraction was the most ideal method,and its parameters was ethanol concentration of 80%,ethanol volume of 100 mL and ultrasound temperature of 45℃,rutin of 11.6% could be achieved by it.There was dose-dependant relation for rutin to clean up free radicle O2-·,and the scavenging index of IC50 was 0.030 5 g/L,which is significant differences exist(t=2.93,P0.05).

    Archaea is one of the most popular research,it not only could survive under extreme environment,such as high temperature,high salinity,strong acid,strong base and anoxic environments,but also exist in various ecological environments.With the huge abundance and the variety of metabolic product,archaea is playing a highly important role in the biogeochemistry processes in the world the study of archaea is important to elucidate the basic laws of life and movement,revealing the interaction between biology and environment.In the article,Methanoarchaea in rumen was introduced,and its diversity also was discussed based on the analysis of 16S rDNA,which has been applied to anaylze more extreme environments of marine archaea.Nowadays,16S rDNA technology is being expanded in the rumen archaeal research,and 16S rDNA gene library of methanogenic archaea was established to study its diversity of rumen archaea.

    Different breeds of meat sheep fed under different stabling conditions,tissue biopsies was applied to detected sheep abomasum tissue changes in histology.The result showed that the abomasum of Dorset sheep was thickest,the next was the Mongolian sheep's,the thinnest was German Merino sheep's,but there were significant differences among them(P 0.05);In wrinkles of the lower gastric mucosa,Suffolk sheep fold gastric muscle was the thickest and Mongolian sheep is the thinnest;In the wrinkle heights of abomasum Gastric area,there were markedly different among Mongolian sheep,Dorset sheep and Suffolk sheep(P0.01),but there were no significant difference among Mongolian sheep,Dekesaier sheep and Germany Merino sheep(P 0.05);In gastric Helicobacter and Suffolk bottom,there was significantly different between Dekesaier sheep and German Merino sheep(P 0.05).

    Waterfowl are the natural repository of avian influenza virus.To study the pathogenic molecular basis of highly pathogenic avian influenza from waterfow,8 recombinant PBD plasmids and rescued the recombinant virus were constructrd by reverse genetics techniques.The total RNA was extracted from the allantoic fluids of chick embryo with the infection of A/duck/HuBei/49/05 virus,PB2,PB1,PA,HA,NP,NA,M and NS were amplified by RT-PCR respectively,and they were cloned into PBD vector.Moreover,they also were transfected into 293 T cells.The result indicated that the recombinant virus was successfully rescued and named as R-DKHB.

    Death receptor 6(DR6 or TNFRSF21)is a member of TNF receptor superfamily.DR6 is a type-Ⅰtrans-membrane receptor possessing four extra -cellular cysteinerich motifs and a cytoplasmic death domain.DR6 is expressed in most human tissues and abundant transcript can be detected in heart,brain,placenta,pancreas,thymus,lymph node and several non-lymphoid cancer cell lines.DR6 can interact with TRADD and active both JNK and NF-κB.In addition,DR6 inhibits proliferation and activation of T and B cell,and it also involved in inducing neurons cell apoptosis.

    By immunohistochemistry,the CD3 + T lymphocytes and CD4 + and CD8 + T cell subsets in chicken cecal tonsil,including appearance,migration,tissue localization and changes of the number,were determined by applying monoclonal antibodies of CD3,CD4 and CD8.The results showed that CD3 +,CD8 + T lymphocytes first appeared on 11 day-old embryo,while CD4 + T lymphocytes in the 15th day.Changes in distribution,CD3 + cell smainly in the mucosal epithelium,CD4 + cells in the mucosa in the lamina propria,but CD8 + cells in the lamina propria and mucosal epithelium.Changes in the amounts,the number of positive cells were all increased dramatically before the 7th day,and at the 7th day,CD3 + cells significantly decreased in the number,but CD4 +,CD8 + T cells had no significant changes in the number.From the 21st day to the 35th day,CD3 +,CD4 + and CD8 + T lymphocytes in the number increase continuously.The conclusion could be made that the immune function can be enhanced in the early post-hatched chicken duodenum.

    In the article,the comparative study of the bioactivity was carried out between eight yeast strains and four mold strains to screen good strain with high activity.The biomass,the crude protein content,living bacteria number,fermentative fragrance of eight yeast strains were determined,and the biomass and synthesis of the enzymes of four mold strain was measured.The result indicated that yeast of Saccharomyces cerevisiae BC,BD,Candida utilis BY,Candida tropicalis BR and Aspergillus niger H3 had the excellent production performance,which can be selected as experimental strains of probiotics for ruminants.

    The trial was designed with three factors and three levels and the aim was to investigate effect of addition of the different proportion the green fodder on Landes goose growth performance.180 Landes gooses with close body weight at 1 day age were randomly divided into nine groups,there were four repeats in every group and 5 Landes goose in a repeats.The result indicated that the young goose in average day appetite(ADA) was higher than that of the control between the groups of green fodder and non-green fodder,and the difference was marked(P0.05),but average weight again(AWA) and feed conversion rate were no differences among them(P0.05),the effect of ADA was the best one with the ratio of 1:2 between green feed and fined feed.In the middle of growth,these groups in ADA and AWA were different,the optimization condition was green feed and fined feed proportion is(1:2) to(3:4).In the adult,the suitable green feed should be added up to the inhered feed,and it not only can satisfy the need of development at the different stage,but also it is essential to fills and feed for the later period.

    In the study,Wuzhumuqin sheep was selected to tested,the greenhouse feeding effect of daily weight gain of Wuzhumuqin sheep was determined by control trial with the selection of different typical model of raising house,the control group was free-pasturing all the year.The results showed that gestation period of female livestock was fed by house and fined feeding,which could keep increase continuously to a certain degree in the next year,and the yearly weight gain was higher that of the control,the difference was marked(P0.05),meanwhile the female in weight gain also was different at the different stage.The different varieties female livestock in daily weight gain was the same,the weight gain was higher that that of the control,but there was markedly different between female sheep and caprine(P0.05).The result of T test showed that lamb in daily weight gain all was higher that of the control at the different stage.Hence,greenhouse feeding has significance to change traditional raising model.

    The sweet sorghum straw can be used as high energy materials to produce fuel ethanol and its by-product is stillage.The basic composition of stillage from fermentation with different techniques was analyzed.The results showed that crude protein content in stllage was the highest with inoculation of 0.25% bacteria,activated for 2 hours,fermented for 5 days.Therefore,sweet sorghum straw stillage after processed can be used as crude feed for ruminants to improve the comprehensive utilization rate of straw.

    Based on social and economic aspects in Abaga Banner ranches,the present station on family pasture was investigated,and the animal performance of Simmental cattle and grassland production was analyzed with the comparative between the model of feeding in winter and traditional grazing.Body weight,body condition,chest length and body length of Simmental cattle were detected every 3 weeks.The result indicated that all indexes of Simmental cattle with house feeding model in winter was higher than traditional grazing,and the body weight loss was less than that of the control,about 0.217 kg in average body weight increase every day.Moreover,the trial group in production also was higher than that of the control.

    Artemisia anethifolia was employed to study the relationship of salt tolerance with Chlorophyll and Betacyanin contents at the different concentration of NaCl.The results showed that Chlorophyll in seedling of A.anethifolia took the tendency of low-high-low as the increase of salt concentration,and Betacyanin contents in beet had an obvious tendency.It was concluded that the content of Chlorophyll in A.anethifolia and Betacyanin in beet could be used as important appraisal indexes to salt tolerance of A.anethifolia.

    This experiment was carried out to assess the growth characterization of tolerance-salt to Saussurea runcinata.The seedling stage of S.runcinata was respectively stressed by NaCl and Na2SO4 at the concentration of(0,60,120,180,240,300 and 360 mmol/L),and these indexes were determined,including root-top ratio,relative growth rate,root length and plant height.The results showed that root-top ratio and root length of S.runcinata were greater than that of the control under salt stress,but the relative growth rate and plant height of S.runcinata were less than that of the control;Root-top ratio of S.runcinata is no significant difference under the same concentration of two kinds salt stress,but the relative growth rate of S.runcinata under NaCl stress is significantly higher than that of Na2SO4 stress.

    In the study,the extraction from stem and leaf of alfalfa was used to investigate the Allelopathic Effect on Germination and Seedling Growth of triticale.The result indicated that the concentration of extract can restrain germination of triticale seed to a certain degree,the inhibition also enhanced as the concentration of extract increased;the weight change in root,stem and leaf of triticale was increased firstly,and then decreased as the concentration of extract increased;when the extraction concentration was 20%,the effect to growth speed and height of triticale was the most significant.

    To accelerate the generation of feed production techniques and improve the nutritional level of livestock fed with grass,7 strains of corn,including Silage 67,Jingang 50,Silage 2,Xingzhu 1,Jinling 7,Jinling 10 and Jinling 14,were introduced by Grassland Working Station of Hangjinhou County of Inner Mongolia in 2009,the comparative trial was carried out.The result indicated that the fresh yield of Xingzhu 1 was the highest one,but it was the lowest in GI;the series of Jinling both were the better in yield and GI,which should be generated at river irrigation district.

    The total-flavonoid in Stellera chamaejasme L.was extracted by ultrasonic technology and its content was determined by colorimetry,and the cleanup rate of the polyphenolic extract towards superoxide anion radical and hydroxyl radical also was determined with control of ascorbic acid.The result showed that the content of total-flavonoid were 24.30%,and the flavonoids extract from S.chamaejsme L.had remarkable function of removing superoxide anion radical and hydroxyl radical in vitro and it was stronger than VC.The conclusion could be made that it is a promising natural antioxidant for the exploitation and development.

    In the study,the relation between oviposition quantity of flat-chest turtle and individual body length,body breadth,body weight was determined,and the exploration on artificial proliferation,cultivation and its reasonable utilization was emphasized.The result showed that with the increase of individual body length,body breadth and body weight,oviposition quantity of flat-chest turtle also increase starting from sexual maturity in the natural multiplication,when individual body length,body breadth and body weight was(25.20±0.26) cm,(17.70±0.33) cm and(1631±27) g respectively,the oviposition quantity attained the expected,however these indexes increase further,the oviposition quantity fall back with lower format.

    In the study,8 week-old ICR mice were selected as the experimental object.The trial animals were fed with different concentration sucrose under long-term stimulation,then they were killed by nitrogen,duodena were obtained,immunohistochemistry method was applied to detect villus shape and the expression level of α-gustducin,meanwhile blood sugar concentration change of mice also was determined intragastric administration before and afterwards.The result showed that rate of duodena velvet rise with the increase of sugar concentration,but there was no significant change in expression of α-gustducin.The conclusion could be made that with stimulation prolongation,the ability of tolerance to sugar enhancement.

    A nutritional beverage was developed by combination of black kerneled rice and milk.Appearance,texture,taste and flavor were chosen as sensory index and the prescription was determined by orthogonal experiment.In addition,the stability also were analyzed by turbiscan technology using six months shelf-life UHT milk as control.The results showed that the prepared sample containing 50.0% milk,5.5% black kerneled rice powder,0.20% emulsifier,4.0%sugar had the best sensory character.For the stabilities of black kerneled rice milk,one compound-stabilizer including 0.20% Microcrystalline Cellulose(MCC),0.15% modified starch and 0.02% к-carrageenan was better than the another one composed of 0.2% MCC and 0.15% modified starch.Moreover,the stabilities of samples were both better than that of the control.Therefore,two above compound-stabilizers can satisfied the need of the stability of black kerneled rice milk in six months shelf-life.

    Many factors take effect on the hatchability rate of eggs,it's important to improve the hatching rate by applying the correct way of laying up eggs and suitable ventilation time.The comparative tests on the way of laying up eggs and ventilation time was made in the study.The result indicated that the fertilization to hatching eggs and hatching rate markedly improved by the project that the small head upwards,the big head downwards,and shift to an earlier time for ventilated time during the process of preservation,the difference is extremely marked(P0.01).

    In the trial,ovocyte of cattle was used as material and trichostatin-A was used as suppressor of de-acetylase.The intensity of Acetylization of histone H4K12 was determined by laser confocal microscopy technique to explore the activity of transacetylase(HAT) and histon de-acylase(HDAC) during reduction division phase of Ⅰand Ⅱ,the treatment concentration of TSA was 80 μmol/L.The result indicated that fluorescence intensity of histone H4K12 couldn't be detected during reduction division phase,and it could be detected from matured ovocytes under the culture condition of TSA,but histone H4K12 disappear completely after the ovocytes were transferred into free-TSA culture medium for 3 hour later.The conclusion could be made that there isn't activity to HAT,but HDAC was activity during reduction division phase of Ⅰand Ⅱ.

    In the study,the aim was to investigate the expression of Hairless gene in Inner Mongolia goat.The degenerate primers are designed based on the known conserved sequence of Hairless gene,after the genome DNA was extracted from blood,then the aim gene was amplified by PCR technique,the product of PCR was sequenced.The result indicated that the gene was about 1 985 bp,including 3 exons and 2 introns,and the result of blast also showed it was specific compared with that of cow and sheep in GenBank.
