彩色小麦成熟胚愈伤组织诱导及再生体系优化

Callus induction and regeneration system optimization of color wheat mature embryos

摘要: 【目的】 研究彩色小麦组织培养再生体系,为建立彩色小麦高频遗传转化体系提供理论基础。【方法】 以中黑麦6号、CM-Z2和中蓝麦1号3个彩色小麦品种(系)的成熟胚作为外植体材料,在愈伤诱导8、14、20 d分别统计诱导情况;采用IAA+KT、IAA+6-BA生长调节剂组合进行彩色小麦愈伤组织的再分化,比较不同处理愈伤组织出愈率、分化率、成苗率等。【结果】 中黑麦6号、CM-Z2、中蓝麦1号外植体在培养14 d时愈伤诱导效果最佳,出愈率在80.54%~95.39%。彩色小麦成熟胚愈伤组织再生能力与基因型、培养基有关,中黑麦6号再生能力最好,分化率在87.68%~97.50%、成苗率在17.26%~61.73%,在含有0.1 mg/L IAA和0.5 mg/L 6-BA、0.5 mg/L IAA和4 mg/L KT的分化培养基中成苗率较高,分别为61.73%、56.25%;其次为CM-Z2,分化率在60.23%~95.45%、成苗率在2.27%~34.09%,在含有0.5 mg/L IAA和4 mg/L KT的分化培养基中成苗率最高,为34.09%。【结论】 中黑麦6号是彩色小麦遗传转化的良好材料;0.1 mg/L IAA和0.5 mg/L 6-BA、0.5 mg/L IAA和4 mg/L KT可以作为彩色小麦中黑麦6号和CM-Z2良好的再生培养基。

Abstract: 【Objective】 To study the color wheat tissue culture regeneration system and provide a theoretical basis for establishing a high-frequency color wheat genetic transformation system. 【Methods】 The mature embryos of three color wheat varieties(lines)of Zhongheimai No.6,CM-Z2 and Zhonglanmai No.1 were used. The induction status was recorded at 8,14,and 20 days after callus induction. Growth regulator IAA+KT and IAA+6-BA combination were used to redifferentiate color wheat callus. The callus rate,differentiation rate and seedling rate of different treatments were evaluated. 【Results】 The explants of Zhongheimai No.6,CM-Z2 and Zhonglanmai No.1 had the best callus induction effect after cultured for 14 days,with callus rates ranging from 80.54% to 95.39%. The regeneration ability of color wheat mature embryo callus was related to genotype and culture medium. Zhongheimai No.6 had the best regeneration ability,with a differentiation rate of 87.68%-97.50%,and a seedling rate of 17.26%-61.73%. The seedling rate was higher in the differentiation medium containing 0.1 mg/L IAA and 0.5 mg/L 6-BA,0.5 mg/L IAA and 4 mg/L KT,with a rate of 61.73% and 56.25% respectively. CM-Z2 followed Zhongheimai No.6,had a differentiation rate of 60.23%-95.45% and seedling rate of 2.27%-34.09%. CM-Z2 had the highest seedling rate in the differentiation medium containing 0.5 mg/L IAA and 4 mg/L KT,reaching 34.09%. 【Conclusion】 Zhongheimai No.6 was an ideal material for color wheat genetic transformation. Regeneration medium containing 0.1 mg/L IAA and 0.5 mg/L 6-BA,0.5 mg/L IAA and 4 mg/L KT were suitable for Zhongheimai No.6 and CM-Z2.
