Study on the Isolation, Identification and Characteristics of Rhodotorula mucilaginosa in Alhagi sparsifolia Silage

Abstract: Alhagi sparsifolia silage was diluted and coated to obtain single bacteria. And single bacteria were purified on PDA medium to obtain three stains of Gram positive bacteria. And the colony morphological identification, physiological and biological properties and ITS (ITS1+5.8S+ITS2)sequencing homology analysis were made on the positive bacteria. The results showed that one strain of red single colony was round or oval,and its centre was ridgy. This stain was able to utilize varied of carbon sources (sucrose, D-galactose, etc.) and two kinds of nitrogen sources [(NH4)2SO4 and L-glutamic acid]. But this strain wasn′t able to utilize KNO3. The whole sequence length of 5.8S rDNA was 616 bp. The homology between this strain and Rhodotorula mucilaginosa ATCC 4054(NCBI accession number:KC881069)was 99.8%by DNA Star Software. So this strain was identified as Rhodotorula mucilaginosa.
