  • Physiological Changes and PLD Gene Differential Expression of Agropyron Cristatum in Osmotic Stress

    For the purpose of investigating drought resistance of Agropyron cristatum,the seedlings were sampled in Osmotic stress conditions for examining the physiological indicators of praline acid, peroxidase (POD),catalase (CAT),malondialdehyde (MDA) and soluble sugar content, and Real-time qPCR analysis of PLD gene expression differences. The results showed that, under Osmotic stress,the changes of proline, POD and soluble sugar content correlated with PLD gene expression significantly on Agropyron cristatum. The research provided physiological understanding for drought resistance and a guide for germplasm screening of A gropyron cristatum.

    The proxidase (POD) and esterase (EST) isozyme characteristics of reciprocal hybrids F1 between Elymus sibiricus and E. purpuraristatus were analyzed by vertical slab polyacrylamide gel electrophoresis technology.The results showed that the reciprocal hybrids F1 had all the bands of both parents at the same developmental stage,and there were significant differences in the number, locus and intensity of the POD and EST isozyme bands among the reciprocal hybrids F1 between Elymus sibiricus and E. purpuraristatus.The isozymes provide evidence for the reliability of reciprocal hybrids F1, and the isozymes can be used for identification of reciprocal hybrids F1 and parental lines of Elymus sibiricus and E. purpuraristatus.

    As one of the major groups of plant growth-promoting bacteria,phosphate-solubilizing bacteria play an important role in the development of green agriculture and control of desertification.The present research mainly focus on phosphate- solubilizing bacteria of farmland,classic grassland and desert ecosystem,however plant growth-promoting rhizobacteria of desert steppe has rarely been studied.The phosphate-soiubilizing bacteria were isolated by the insoluble phosphate medium, and the phosphate-solubilizing ability of the isolates were detected by the potassium persulfate digestion and Mo-Sb anti colorimetric method.42 phosphate-solubilizing bacteria were isolated from the rhizosphere of four dominant plants in desert steppe of Siziwang Banner.The 42 strains were classified into 5 groups and 19 genuses.Among them,19 isolates (45.2%) belong to the Actinobacteria,5 strains (11.9%) belong to the Bacilli,16 strains (38.1%) belong to α-proteobacteria,1 strain (2.4%) belong to β- proteobacteria,and 1 strain (2.4%) belong to γ-proteobacteria.The results showed that strain GP0501,GP0506,GP0507 and GP0509 have the strongest phosphate solubilizing ability,the phosphate solubilization capacity was 77.57,65.89,58.08,68.02g/μ mL,respectively.The 4 strains have great potential of application,which all belong to Bacillus.

    This study used Castor cuhivar Lm-aLmAB2 as object for research inflorescence differentiation of its three types progeny in microscopic level and confirmed the critical transitional period from vegetative growth to reproductive growth. We also provided a reference for ectopically cultivation and breeding of castor in northeast region. The results were as follows: the climacteric of flower bud differentiations in different types were about 5 to 6-leaf stage; castor inflorescence was panicle,the tight arrangement of floret followed by bisexual line, isofemale line,female line;the diameter of inflorescence in bisexual line was largest than isofemale line and female line;flower was incomplete;the specificity salix-shaped leaf occurred at 7-leaf.

    In order to further clear the photosynthetic characteristics and production mechanism of the different effect of drip irrigation,during 2012 and 2013,this experiment by spring maize as material,effects of drip irrigation for comparison, studied the four kinds of effect of drip irrigation on maize net photosynthetic rate (Pn), transpiration rate (Tr),stomatal conductance (Gs), intercellular CO2 concentration,leaf instantaneous water use efficiency (WUEi),SPAD and fluorescence parameters and the influence of the grain yield. Results indicated that the QM,KM,SM,JM treatment of Pn,Tr and Gs,WUEi,SPAD,fluorescence parameters and grain yield were significantly higher than that of CK, it showed very significant positive correlation, 4 drip irrigation mode of yield increased by 7.6%~28.5% than CK, one of the biggest increase rate of QM, increased by 28.5% and 18.3% than CK, followed by KM, increased 24.0% and 12.2% than CK. It was the best way of drip irrigation in this area.

    The simple identification method for the uninucleate stage of processing tomato microspore.Carmine acetate dyeing, Neutral red,Fuchsin basic,Carbol fuchsin were used to stain the cell nuclear of microspore in uninucleate stage.Results showed that Carbol fuchsin was the best identification method for the developmental stages of processing tomato microspore.
    Effects of Different Irrigation Quantity on Photosynthetic Physiology and Water Use Efficiency of Cucumber in Greenhouse

    The experiment was studied to improve water use efficiency of cucumber in greenhouse and explore cultural model with economizing water. As the contrast of irrigation amount in experience of farmer, the cucumber were tested as the experimental materials to study the effect of controlling irrigation quantity on photosynthetic physiology, yield and water use efficiency of grafted cucumber and self-rooted cucumber in the greenhouse. The results showed that as follows: there were obvious effects of appropriately reducing irrigation quantity to promote photosynthesis and improve water use efficiency. The treatment of I1 was significantly more than the treatment of I2 in photosynthesis and chlorophyll content of cucumber. Compared with self-rooted cucumber, grafted cucumber had better performance in photosynthesis, yield, and water use efficiency. In the treatment of II,yield and water use efficiency of grafted cucumber respectively improved 7.16%,compared with self-rooted cucumber; In the treatment of I2,yield and water use efficiency of grafted cucumber respectively improved 7.83%,compared with self-rooted cucumber. On the whole, in this experiment, the treatment of I1-G had best performance in the hand of photosynthesis, yield, and water use efficiency, was a better cultivating and irrigation mode, and had a promotional value.
    Comparison on the Colonization of Leymus chinense Arbuscular Mycorrhizal in Different Salinities

    The relationship between Leymus chinense and AM (arbuscular mycorrhizal) fungi in different salinities of Songnen alkaline grassland was investigated in this study, the roots of Leymus chinense from low salinity, middle salinity and high salinity were gathered respectively. Using microscopy techniques, arbuscular, vesicles,and other forms of mycorrhizal structure type were observed after Trypan-blue staining and producer. Results showed that AM fungi colonized all Leymus chinense in different salinitie, among which middle salinity got the highest colonization rate, infection intensity, vesicular abundance and arbuscular abundance of 100%,31.78% and 76.25%,while high salinity got the lowest colonization rate, infection intensity, vesicular abundance and arbuscular abundance of only 90%,27.15%,16.90% and 57.74%.There were also significant differences on the colonization of Leymus chinense arbuscular mycorrhizal in the same salinity (P〈0.05). The AM type of Leymus chinense was A-type in low salinity and middle salinity, while the AM type of high salinity was P-type.
    Population Diversity and Phylogeny of the Cultivable Bacteria in the Surface Water of East Juyanhai Lake

    East Juyanhai as one of the major lakes in Inner Mongolia Plateau, the survival status of microorganisms in water is not clear.In this study,74 bacterias was isolated from lake water samples. According to the 16S rDNA sequences system phylogenetic analysis showed that, Alphaproteobacteria strains up to a total of 39 Accounts for about 53% of the total bacteria; Actinomycetes (Actinobacteria) is the second dominant groups,a total of 19 strains,representing 26% of all strains.ln addition, the nine isolated from Bacillus groups,six from γ-proteobacteria groups and one from the β-proteobacteria (Betaproteobacteria) groups. These bacteria belonging to 23 genera, which Caulobacterales of Brevundimonas dominant population, a total of nine.

    Through the "3414" completely plan of field experiment, the best dosage of farmyard manure, nitrogen and phosphorus fertilizer applied in planting winter wheat in the south of Ningxia semi-arid gully area was discussed in this paper. Using shyfxExcel software analysis of the mathematical model of the winter wheat production target, it showed that nitrogen was the main factor restricting output. Preliminary concluded that gully area heilu soil (mengyuan heilu soil) of winter wheat yield was more than 300kg/667m2, the dosage of farmyard manure 2500kg/667m2, N11.6 666.7 kg/m2, P2Os10.4kg/667m2; When the maximum yield of 319.8kg/667m2, the dosage of farmyard manure 2900kg/667m2, N13.7kg/667m2, P20512. 4kg/667m2; when the best yield at 297.4kg/667m2, the dosage of farmyard manure 2000kg/667m2,N10.5kg/667m2,P2052.5kg/667m2;Nitrogen basal fertilizer and topdressing fertilized 50% each, farmyard manure should be widely applied in agricultural production in the future, and nitrogen and phosphorus fertilizer used should adjust to local conditions.

    In order to facilitate the adjustment of agricultural structure, and lead farmers to scientific fertilization,Zalantun had truly mastered the basis of production capacity on the local arable lands through the project of formula fertilization by soil testing, to provide a strong basis for scientific fertihzation. The changes and distributions of organic matter,a large number of elements and medium trace elements to the existing problems and fertilization among farmers and the following suggestions were put forward in this paper, and other suggestions of increasing organic fertilizer, fertilizing soil fertility,scieutific application of medium trace elements and promoting formula fertilization by soil testing.

    Using different dosage gradient test of micronutrient fertilizer, the effect of micronutrient fertilizer on potato yield and agronomic traits was studied, the results showed that with the micronutrient fertilizer dosage increased, potato number of per plant, big potato number of per plant, yield of per plant, yield of 667m2 were all increased, when micronutrient fertilizer dosage of 600 g/600 m2, the above index all reached maximum; increased more micronutrient fertilizer dosage, the all parameter values were showed declined trend.

    Using the annual average temperature, maximum and minimum temperature data during the year 1959-2013 of the Kezuohou Banner station, the correlation analysis showed that, the climate changed significantly, and the rising range of the minimum temperatures are obviously higher than that of maximum temperatures, it showed that the warming of day and night was asymmetry and led to decrease the diurnal temperature range. The temperature warming in winter is finest achievement in the annual temperature variation. Since the the climate warming began in 1960s, it had been exceed the significant level in the early of 1980s.The response of maximum temperatures to warm was far behind the minimum temperatures.

    In this paper, the Yakeshi National Meteorological Station 1961-2010 and 50 years daily average temperature by 0~, 5~C stable,10℃ accumulated temperature data was analyzed. Study on the interannual variation, temperature change in the early and final day change trend of accumulated temperature, it showed that Yakeshi accumulated temperature had an increasing trend, which tend to stabilize the average rate of 0℃ was 74.6℃/10a,5℃ tendency for the average rate of 67.5℃/10a,10% tendency for the average rate of 69.4℃/10a.And the value of accumulated temperature, the days of accumulated temperature, the number of days at the beginning of the final date showed fluctuating upward (or delay) trend.

    The average temperature, highest temperature, lowest temperature of climate trends and characteristics in Jining area from 1971-2010 was analyzed in this paper, the results showed that the warming was significant between 40 years, the rate of average temperature increasing more than the national and Inner Mongolia average level, and showed the asymmetric warming, the lowest temperature increasing early at high rate, and mutation analysis also proved that. In addition, through the wavelet coefficients of real component analysis, the average temperature and the highest and lowest temperature changes in Jining area of nearly 40 years were mainly at tO years cyclical swings.

    There existing household biogas, small farming community and joint household biogas project and Large biogas in XilinGol League rural and pastoral, village and county biogas service system framework has been erected,the project has achieved remarkable economic,social and ecological benefits.But there are some problems, for example, digesters utilization was not high (10%-20% in the coldest season),property service outlets were not fully appropriate effect (sample survey only 44.4%), biogas utilization level was low,the construction of large biogas was slowly.Analysising the reasons and proposing the next priority:Active in biogas utilization tests and demonstrations,Efforts to explore innovative biogas construction management and post-the service mechanism,Increased guidance and inspection efforts to strengthen project management,introducing commercialization digesters in actively and steadily.Suggestions: Increasing investment in central special funds,giving appropriate wage subsidies to biogas service outlets managers,introducing large biogas project supervisions and management measures and muhi-energy complements as soon.
