  • Identification of Swine-derived and Bovine-derived Materials in Animal Products by Multiplex Fluorescent PCR Assay

    [Objective ] To develop a multiplex fluorescent PCR assay method to detect swine-derived and bovine-derived materials in animal products. [ Method ] Two pairs of specific primers and two fluorescent-labled probes were designed and synthesized according to mitochondrial DNA gene sequences. The multiplex fluorescent PCR reaction system was established through the optimized screening of reaction system and reaction conditions, and swine-derived and bovine-derived materials in animal products were detected in the same fluorescent PCR reaction system. Finally, the sensitivity and specificity of the assay were evaluated. [ Result ] The method successfully detected swine-derived and bovine-derived materials from 16 animals. Swine-derived and bovine-derived materials could obtain responsible S-type amplification curve, while there was no amplification curve from the other 14 animals. The detection limits of the assay for swine and bovine were either up to 10^-5, which was consistent with that of single-weight fluorescent PCR assay method. [Conclusion ] The multiplex fluorescent PCR developed in the study has strong specificity and good sensitivity, so it is suitable for the detection of swine-derived and bovine-derived materials in feed, meat products, milk products, etc..

    [Objective]To establish a determination method of Zn in fish by digestion tank digestion and flame atomic absorption spectrometry. [Method]The fish was dissolved by digestion tank digestion, and then Zn in fish was determined by flame atomic absorption spectrometry. [Result]This method had good linear relation when the concentration range of Zn was 0-2.0 mg/L, and the correlation coefficient was 0.999 7. The detection limit of Zn was 0.002mg/mL, with the relative standard deviation of 3.12%. The recovery of Zn was 95.0%-102.5%. [ Conclusion ]The method is simole, raoid and accurate, so it is suitable for the determination of Zn in fish.
    Comparison on Multiplication Effect of Canine Distemper Virus in MA-104, Marc-145, and Vero Cells

    After canine distemper virus (CDV-XN 112 strain) was inoculated into MA- 104, Marc- 145 and Vero cells, the sensitivity of three types of monkey kidney to CDV was compared by observing cell pathological effect, determining the time for harvesting virus and comparing virus titers. The results showed that three cells were suitable for CDV, with typical cell pathological effect, and the titers of virus harvest fluids were more than 104.5 TCID50/0. lmL, among them Vero cell was more cost-effective and ideal for proliferation CDV.
    Detection of Dust in Scale Chicken Farm

    Excessive dust concentration in chicken house is hazardous for workers' health and poultry productivity, and also is harmful to the environment. To understand dust spatial distribution is helpful to analyze the emission and transmission of dust, and is helpful to develop more effective control methods for dust. The content of dust in the door and central of chicken house in scale chicken farm throughout the year was detected. Results show that the full dust of central bit point had significant difference between spring and winter as well as between autumn and winter (P〈0.05), and had very significant difference between summer and winter (P~0.01). The respirable dust PM10 had significant difference between summer and autumn (P〈O.05), and had very significant difference between spring and winter as well as between autumn and winter (P〈0.01). The seasons respirable dust PM10 in central had no significant difference with that in door(P 〉0.05 ). The full dust TSP in central also had no significant difference with that in door(P 〉0.05).

    Based on climate condition in Yanchi County, six maize varieties were introduced in the test. The result showed that, there were no significant differences between Tunyu No.1, Zhengdan 958, Zhongdan 5485 and control group, and adapt to local growing conditions, so they could be regarded as the spreading variety in the area. Although Danyu No.3 profit high, air-dry moisture content was 74.3%, which meet the requirement of silage in the growth period, so it could be regarded as the silage crop spreading variety in the area.

    In order to choose the best corn variety in Yanchi area, Liaoyuan No. 1, Keduo No.4, Zhongyuandan No.32 and Liaoyuan No.1 F3 were compared with Zhongdan No.2. The result showed that, Liaoyuan No.1 and Zhongyuandan No.32 were high resistant to diseases, resistant to insects, enduring to drought, cold-resistance and drought tolerance, so it could be regarded as the spreading variety in the area.
    Effects of Different Years of Birth on the Birth Weight of Zhongwei Goat Lamb under Stabling Conditions

    The birth weight of Zhongwei goat lamb under stabling conditions from 2004 to 2010 was analyzed. The results showed that the birth weight of lamb from 2004 to 2010 was very close and the difference was not significant(P 〉0.05 ). The birth weight of male lamb in 2007 increased by 12.87%, 12.04% ,3.37%, 10.83%, 10.04% and 10.83% than other years respectively. And the birth weight of female lamb in 2007 increased by 8.88%, 11.02%, 1.43%, 5.22%, 11.46% and 2.55% than other years respectively. The birth weight of male lamb in the same year was higher than that of female lamb, without significant differences (P 〉0.05 ). The birth weight of male lamb during 2004-2010 was increased by 5.02%, 7.87%, 6.84%, 8.87%, 3.36%, 10.27% and 0.70% than that of female lamb.

    G -protein mediated signaling system is the most common signal transmission in cells. G-protein coupled of signal transmission system consist of G-proteln coupled receptor (G-protein coupled receptors, namely as GPCRs), G-protein and effector. In 1977, Ross and Gilman confirmed G-protein could be used as cell receptor and effector between the converter. And a large number of studies found that the G-protein coupled signal conduction system existed in plants and animals widely. The role of G-protein and GPCRs in the cell conduction was mainly discussed.

    Mealies alcohol grains in wet state is protein feed and by-product of grain distillery.In the United States, 60% of mealies alcohol grains were dry condition DDGS which used for domestic consumption and export; the other is wet state which used as feed. In our country, most of production and consumption was mealies alcohol grains with dry condition, which means that wet state ones had huge market. The production process, nutritional components, feed value of mealies alcohol grains were described, utilization prospect of mealies alcohol grains in wet state was analyzed.

    Studied contents of alfalfa saponins of Medicago sativa, Medicagofalcata and Caoyuan No.2 in different breeding period. The variance analysis was made by using program of SPSS 11.5 software, and multiple comparison of was carried out with Duncan methods. The results indicated that from regreening stage to early blooming time, the contents of total saponins in plants were : Medicago sativa〉 Caoyuan No.2 〉 Medicagofalcata. The contents of total saponins in lucerne was noticeably higher than Medicagofalcata (P〈0.01). At full bloom stage and pod-besring period, the contents of total saponins in Caoyuan No.2 was higher than the other varieties. At the maturation stage, the contents of total saponins in Medicago falcata was higher than the other varieties.The contents of total saponins plants of Medicago sativa and Caoyuan No.2 in pod-besring period were noticeably higher than other growing periods (P〈0.05). At full bloom stage, the contents of total saponins Medicagofalcata was higher than the other growing periods.

    [Objective] The control effect of piglet early weaning diarrhea (PEWD) by adding glucose oxidase and phytohemagglutinin to diet was studied to explore the effect on performance and intestinal health of piglet. [Method] One hundred and twenty weaned piglets were randomly divided into three groups, that was controlled, group I and group 11. Under the uniform raising management pattern, piglets in control were fed on routine basal diet; piglets in group I were fed on diet with 0.5% glucose oxidase; piglets in group II were fed on diet with 0.5% glucose oxidase and 0.1% phytohemagglutinin. The diarrhea frequency, feed intake, death number and empty body weight per piglet in the beginning and at the end of the experiment were recorded every day. The average daily gain, diarrhea rate, average daily feed intake, feed conversion rate and mortality were calculated. [Result] Compared with control, the average daily gain of piglets in group I and II were significantly improved by 25.0% and 26.8% respectively(P〈0.01 ); the feed conversion ratio of piglets in group I and I1 were significantly decreased by 17.2% and 18.7% respectively(P〈0.01 ); the diarrhea rate of piglets in group I and II were significantly decreased by 56.0% and 65.5% respectively (P〈0.01); the survival rates of piglets in group I and lI were improved by 7.4% (P〉0.05)and 13% (P〈0.01), respectively. [Conclusion] Adding glucose oxidase and phytohemagglutinin to diet could improve feed conversion rate and piglet survival rate and obviously reduce piglet diarrhea rate.

    The pathogenetic milk samples from a cattle-raising community in Tumoteyou County of Baotou were selected to perform tests for pathogen isolation and culturing,smear staining,biochemical identification,drug sensitivity to find out which type of mastitis pathogenic bacteria the cows had and which kind of drugs they were sensitive to. The results showed that the prevalence of cow mastitis in the community is caused by Escherichia coli,Streptococcus agalactiae and Staphylococcus aureus. The identified pathogens had certain drug-resistance to long-term soil enzyme and the sulfate chains of enzyme, but they were extremely sensitive to cephalexin and were also highly sensitive to the new enzyme and ampicillin sodium.
