  • Chromosome Ploidy Determination of 9 Populations of Agropyron mongolicum Keng by Flow Cytometry

    Three kinds of separation medium were used to prepare nuclear suspension of Triticum urartu. The preparation effect of Otto buffer was the best. Otto buffer was selected to prepare single cell suspension of 9 populations of Agropyron mongolicum Keng. The number of cells was counted by a microscope and its chromosome ploidy was detected by using flow cytometry (FCM) with T. urartu diploid as control. The results showed that the results detected by flow cytometry were consistent with the results by traditional chromosome preparation method and they were diploid.
    Effects of IBRV Infection on the Nitric Oxide Produced by Bovine Monocyte Macrophage

    To discuss the immune mechanism of infectious bovine rhinotracheitis virus (IBRV) after infecting on bovine monocyte macrophages, bovine peripheral blood mononuclear cells were isolated by Ficoll-Hypaque density gradient centrifugation and monocyte macrophages were obtained by adherent culture. The content of nitric oxide (NO)secreted by bovine monocyte macrophages was detected by Griess method after IBRV was infected 6, 12, 24 and 48 hours. The results showed that the content of nitric oxide (NO) secreted by bovine monocyte macrophages after IBRV was injected was higher than that of control group (P〈 0.05) and the amount of nitric oxide produced by macrophages reached the highest level (607.87μmol/L)after 24 hours. IBRV infection could affect the amount of nitric oxide secreted .by monocyte macrophages,which might be related to pathogenesis caused by IBRV.

    In order to find out a fast, simple and reliable detecting method, the content of K in potato was detected by using flame method and TAS-986 atomic absorption spectrophotometer. Results showed that the correlation coefficient (R2) of standard curve was 0.990 0, and the average recovery was 91.58%, and the precision and repeatability (RSD) was less than 10%. Therefore, the determination method of K content in potato had some advantages of reliability, high precision, good repeatability and high recovery.

    In order to obtain transgenic plants of alfalfa with stronger drought resistance ,dehydrin gene of A mmopiptanthus mongolicus was transformed into the genome of Zhongmu No. 2 mediated by Agrobacterium tumefaciens with alfalfa variety Zhongmu No. 2 as gene transformation receptor. The results showed that the induction frequency of cotyledon and hypocotyls as explant calli was the highest and the best co-culture time for explants and A. tumefaciens was 5 days. 126 resistant plants were obtained. After the identification by PCR and RT-PCR, 11 positive transgenic alfalfa plants were obtained. The elementary phenotype analysis and proline analysis after drought treatment indicated that transgenic alfalfa had stronger drought resistance ability.
    Comparative Study on the Epidemic Prevention Effects of Different Drugs on Zhenning Yellow Chicken in Brooding Period

    Four different drugs were used to make a disease prevention experiment on 2 000 chicks of Zhenning Yellow Chicken for 8 weeks. The results showed that the survival rate of chicks of Zhenning Yellow Chicken was more than 94%. The disease prevention effects in test Ⅲ group was the best and the survival rate was over 97%. The disease prevention procedure could be popularized and applied in practical production of peasant households.
    Study on the External Morphological Differences of Tylototriton shanjing in Mojiang and Tengchong

    The shape structure of adult Tylototriton shanjing collected from Mojiang and Tengchong were compared under laboratory feeding conditions from May of 2011 to March of 2013 . The results showed that Tengchong individuals had blacker color, Mojiang individuals had saffron yellow skin. The average body length of Tengchong individuals and Mojiang individuals were 151.80 mm and 134.32 mm respectively. The ratio of tail length to body length of Tengchong individuals and Mojiang individuals were 0.489 and 0.479 respectively. There were irregular black knobby on the back and body side of Tengchong individuals and Mojiang individuals. The knobby size and shape on the both sides of back in Tengchong individuals were the same, while the knobby of Mojiang individuals had irregular shape and different size. And the knobby of Mojiang individuals was more intensive and remarkable than Tengchong individuals. As for the secondary sexual characteristics, the average cloaca length of male Tengchong individuals was 8.2 mm and that of male Mojiang individuals was 7.7 mm.
    Effects of Different Hormones on Estrus Induction and Farrowing Effect of Bitches

    In order to study effects of different hormones on estrus induction and farrowing effect of bitches, 110 bitches without estrus for a along time were divided into five groups, among them group Ⅰ was treated with intramuscular injection of PMSG; group Ⅱ was treated with intramuscular injection of the combination of PMSG and HCG; group Ⅲ was treated with intramuscular injection of the combination of PMSG and LRH-3; group IV was treated with intramuscular injection of the combination of PMSG, PGF2α, E2 and LRH-3; group V was control group. Results showed that, as for estrus, group IV had the best effect, and showed high reaction in degree of vulva swelling, flowing-blood secretion and keratinization degree of vaginal epithelium; the estrus rate of group Ⅳ was 32%, 28% and 20% more than that of group Ⅰ, group Ⅱ and group Ⅲ respectively (P〈0.01). Group IV had the highest conception rate of 76% which was 35%, 20.4% and 10% more than that of group Ⅰ, group Ⅱ and group Ⅱ respectively; group Ⅳ had very significantly difference with group Ⅰ and group Ⅰ, and had significantly difference with group Ⅲ. The litter size of group IV was 130 which was 92, 74 and 45 more than that of group Ⅰ , group Ⅱ and group Ⅲ, respectively (P〈0.01). Control group wasn't treated with any drug, and it had the lowest estrus rate (10%), and its conception rate was 0. It is suggested that the combination of PMSG, PGF2ct, E2 and LRH-3 had the best effect of estrus induction, and both conception rate and litter size were better than those of single use or the other combinations.
    Effects of Probiotics on the Intestinal Flora of Diarrhea Rats Induced by Antibiotics

    [Objective] The research aimed to evaluate the effects of probiotics on intestinal flora of diarrhea rats induced by antibiotic. [Method] The diarrhea model of rats was build by cefiriaxone. The rats were divided into groups. Except control group, rats in other groups were injected by gavage with ceftriaxone for four clays to build the diarrhea model of rats. Then rats were treated with probiotic preparation for 7 days. The indicators of rats were detected. [ Result ] After they were treated one week, the body weight of young rats rose up. And lactobaeillus and bifidobacterium recovered to control level, but they were still better than natural recovery group. [Conclusion] The probiotics could improve antibiotic-induced diarrhea in rats and have certain role for flora imbalance.
    Comparison on the Degradation Law of Corn Silage from Xinjiang and Inner Mongolia in Sheep's Rumen

    In order to understand the degradation rate of the western silage, some kinds of corn silage were selected from Xinjiang and Inner Mongolia, and effective degradation rates of some nutritional components including dry material (DM), crude protein (CP), neutral detergent fiber (NDF) and acid detergent fiber (ADF) in the corn silage were determined by using the method of nylon bag technique so as to apply a reference for improving the utilization of corn silage in the production of the western mutton sheep. Results showed that the nutritive value of corn silage in Xinjiang was higher than that in Inner Mongolia, and the effective degradation rates of DM, CP, NDF and ADF of corn silage in Xinjiang were higher than those in Inner Mongolia, which suggested that the corn silage in Xinjiang was easier than that in Inner Mongolia to degrade in sheep's rumen.

    [ Objective ] In order to understand the bacterial pollution level of beef feed from cattle farms that located in two regions in Xinjiang, the Coliform group and Salmonella in 18 collected feed samples were detected. [Method] The method of Salmonella detection was in accordance with provisions of the national standard GB/T 13091-2002, and the method of Coliform group detection was in accordance with provisions of the national standard GB/T 18869-2002. [ Result ] The straw feed had the highest number of Coliform group, which was up to 11 000/100 g; However, processed feeds like pellet feed, cottonseed feed and rapeseed feed, the number of Coliform group were all lower than 40/100 g; and there's no Salmonella to be detected in all feed samples. [ Conclusion ] All the 18 detected beef feed samples were contaminated by Coliform group at different degrees.
    Effects of Different Nutritive Regulating Agents on Production Performance of Holstein Bull in the Latter Finishing Period

    Sixty Holstein hulls of about 450 kg were divided into three groups according to weight and types, among them group one and two were fed with different nutritive regulating agents and uniform basal diet, and group three (control group) were only fed with basal diet. Group one was fed with nutritive regulating agent A at dosage of 100 g/d, and group two was fed with nutritive regulating agent B at dosage of 100 g/d. Results showed that after the group one and two were fed with different nutritive regulating agents and uniform basal diet, their average daily gains were 2.10 and 2.28 kg, respectively, among them the average daily gain of group two was significantly higher than that of control group; the value of weight gain, fattening income and daily average income of group one and two were all obviously higher than that of control group, and group two had the highest economic benefit.

    Soybean has abundant fresh nutrients such as soy isoflavones, hydrolyzed soy protein and soy lecithin, which lasted skin moisturizing, tender and smooth. So soybean is safe and reliable high-quality raw materials of original ecological skin care products. The main components of aloe are anthraquinone complex and it is bitter and cold. Aloe has a catharsis effect and it can regulate the physiological functions of skin. So aloe is one of the best skin care products. The results showed that the mixture of soybean and aloe had better skin care effects than general skin care products.

    The development of prolific meat sheep industry is a new increase point . 110 demonstrate fields (households) for developing prolific meat sheep were established in Hami Area of Xinjiang in 2012. The development status, existing technique problems of prolific meat sheep breeding industry in two counties and a city of Hami were investigated. The production status of main varieties, the sheep number in stock in the distributed area, reproduction performance, epidemic prevalence laws and assessment of production benefits were mastered.     Prevalence Situations and Change Trend of Brucellosis in Xinjiang

    In order to master the prevalence situations and change trend of brucellosis in Xinjiang, based on collecting and cleaning the monitoring data of brucellosis in this area, the monitoring data of brucellosis in 96 counties (cities, districts) of 14 areas(prefectures, cities) in Xinjiang were preliminarily analyzed during 2005-2011. And the prevalence and epidemic situations of brucellosis distributed in various kinds of animals and different regions were compared. The results showed that the average positive rate of cattle brucellosis and sheep brucellosis during 2005-2011 were 0.66% and 0.61% respectively. And the average positive rate of cattle brucellosis in large-scale farm and administrative villages were 0.37% and 0.77% respectively. The average positive rate of sheep brucellosis in large-scale farm and administrative villages were 0.77% and 0.52% respectively. The positive rate of cattle brucellosis in Taeheng was the highest, and the positive rate was 2.91%. The positive rate of sheep brucellosis in Urumehi was the highest, and the positive rate was 14.67%.
