绵羊miRNA-1185-5p的基因组定位、靶基因预测和功能分析

Genomic Location, Target Genes Prediction and Their Function Analysis of miRNA-1185-5p in Sheep

摘要: miRNAs是一类小分子非编码RNAs,在机体许多生理及病理过程中发挥调控作用。该课题组前期研究发现,miRNA-1185-5p在经产双羔母羊卵巢中表达水平显著低于经产单羔母羊。为进一步深入挖掘miRNA-1185-5p在绵羊生殖过程中的作用,利用生物信息学手段分析了miRNA-1185-5p的基因组定位,预测其靶基因,并对靶基因进行GO功能分析。结果表明,miRNA-1185-5p是位于绵羊18号染色体的内含子miRNA;利用miRDB和microT 2种方法预测获得miRNA-1185-5p的23个靶基因,这些靶基因主要参与细胞增殖、分化和凋亡过程以及代谢等生物学过程,其中,靶基因AhR直接参与生殖过程。该研究说明miRNA-1185-5p可能通过作用于AhR等靶基因对绵羊生殖过程发挥调控作用。

Abstract: miRNAs are a class of small molecule non-coding RNAs that play a regulatory role in many physiological and pathological processes of the body. Our previous research found that the expression level of miRNA-1185-5p in the ovary of double-lambing ewes was significantly lower than that of the single-lambing ewes. In order to further explore the role of miRNA-1185-5p in reproductive process of ewes, the genomic location of miRNA-1185-5p was assessed by using bioinformatical methods. Furthermore, the target genes prediction and their corresponding GO annotation of miRNA-1185-5p were bioinformatically performed. The results showed that miRNA-1185-5p was an intron miRNA located on sheep chromosome 18; a total of 23 genes were predicted as the target genes of miRNA-1185-5p via miRDB and microT methods, which were involved in cell proliferation, differentiation, apoptosis, metabolism and other biological processes. Among the target genes, AhR gene was found to directly participate in the reproductive process. This study suggests that miRNA-1185-5p may regulate the reproductive process of ewes by acting on its target genes such as AhR.
