  • Orthogonal Design and Process Optimization of General Flavone Extraction from Chrysanthemum indicum L.

    [Objective] To deeply study extraction process of the general flavone which is active antibiosis ingredient from Chrysanthemum indicum L. [Method] Orthogonal method was used to design extraction experiment. Ethanol concentration, extraction time and temperature were set as influential factors. The general flavone content was measured by absorbance method. Influence degrees of these influential factors on general flavone content in extraction liquid were discussed. [ Result ] According to orthogonal experiment results, it was thought that extracting time and temperature were major influential factors, and ethanol concentration was minor influential factor. [Conclusion] It was determined that the best ethanol concentration was 30%, extraction time was 45 min, and extraction temperature was 70 *C.

    Three groups of mice were given with swainsonine (SW), thermopsine (Ts) and their mixtures by gavage, respectively Four mice from each group were randomly chosen to take the anticoagulant blood of the eyes after 7, 14, 21, 28 and 35 days respectively. The peripheral blood T cell subsets CD4+ and CD8+ were assayed by flow cytometry, and the ratio of CD4+ to CD8+ wa calculated. The results showed that the low dose of SW could play a significant role in enhancing immunity of mice, while th, medium and high concentration SW played an active role in short time, but decreased immunity along with the extension of time the low concentration of Ts didn't work on immunity in mice, while the medium and high concentration of Ts could reduc, immunity; the low concentration of mixtures had little effect on the immunity of mice, while the high concentration of mixture could still decrease immunity significantly.

    The growth change of follicular of thyroid gland was detected by using tissue method. Results showed that the follicular of thyroid gland became larger with the increase of days, and the height of follicular epithelial cell at 30 days old was obviously higher than those of the others. The study on the ultrastructure of follicular epithelial cell at 1, 10 and 30 days old showed that the function of epithelial cell was in vigorous stage, while the function of ollicular epithelial cell at 60-90 days old was in relatively stationary state.

    [ Objective ] In order to understand the ambient air quality of beef cattle cowshed in early winter of Aksu, Xinjiang, an evaluate ambient air quality in beef cattle cowshed. [Method] Four kinds of main harmful gas (CO2, NH3, CH4 and H2S) in cowshed of beef cattle farm and PM10 and TSP in ambient air in Aksu of Xinjiang were measured. Ambient air quality in cowshe was evaluated. [Result] Measurement result showed that CO2 concentration in beef cattle cowshed was 404 mg/m3,NH3 concentration was 4.0 mg/m3, CH4 and H2S were less than 0 mg/m3, PM10 was 0.04 mg/m3, and TSP was 1.222 mg/m3. The highest and daily average concentrations of CO2, NH3, CH4, H2S, PM10 and TSP were all lower than ambient air quality standard of the livestock and poultry. [Conclusionl The air quality of beef cattle cowshed belonged to clean level, in accord with environmental standards of the beef cattle breeding.

    Three family ranches of Elitu in Zhengxiangbai Banner of Xilinguole were investigated for researching balance of forage and livestock in 2011. The results showed that three family ranches were overgrazing with stocking rates of 1.01, 0.73 and 1.17 head/hm2, respectively. In response to above situation, stocking rates were adjusted by using grassland and livestock energy balance model. No.1 family ranch should eliminate all wethers and 12.5% of ewes, and the stocking rate was adjusted to 0.82 head/bin2; No.2 family ranch should eliminate all wethers and 15.0% of ewes, and the stocking rate was adjusted to 0.50 head/hm2, meanwhile species should be improved;No. 3 family ranch should eliminate all wethers and 28.8% of ewes, and the stocking rate was adjusted to 0.77 head/hm2. This model should be benefit for increasing farmers' income.

    Transgenic process may be with some undesirable molecules, such as error-connected carrier, DNA fragments in the PCR process, vector without inserted DNA fragment, other recombinant DNA molecule and insert incorrect fragment. These will bring some troubles to transgenic cloned. Studies showed that some carrier may carry different resistance-detecting genes, and the require cloning and unwanted non-purpose cloning region could be separated through a suitable screening step to achieve the object of purified DNA. The test gene and its principle of three carriers were described.

    Wet lees fermenting protein feed is prepared through that the corn is treated through yeast fermentation, extraction alcohol, probiotics fermentation and sealing packaging. The power feed of hens was added with wet lees fermenting protein feed, and then the control effects on ammonia concentration of chicken house and egg yield of laying hens were studied. The results showed that if the power feed of hens was added with 15% of wet lees fermenting protein feed, the machinery feeding wasn't affect, and there was no mildew and denaturalization phenomenon, and the feed intake of hens increased. Compared with control, the egg yield of tested group was increased by 5.2% with an increase of 6.6 kg, and the economic benefit of tested group was increased by about 10% with an increase of 51.7 yuan. The ammonia concentration of chicken house decreased largely, and it was 22.8 mg/L less than control, which was decreased by 32.0%.
    Effects of Compound Probiotics on Growth Performance and Immune Function of Broilers

    Compound probiotics product was added in feed to study the effects on growth performance and immune function of broilers. One hundred and fifty one-day-old AA broiler chickens were randomly divided into 2 groups with 3 replicates in each group and 25 chickens per replicate. The pre-trial period lasted for 4 days,and the experiment lasted for 36 days. The results showed as follows: compared with control, the feed-meat ratios of tested group at the age of 12, 19, 26 and 40 days all decreased significantly (P〈0.05), and the feed-meat ratio of tested group was lower 10.9% than that of control in the whole test cycle sifnificantly (P〈0.05). The spleen index of tested group at the age of 33 days was higher 55.1% than that of control (P〈0.05), and the Bursa index of tested group at the age of 26 days was higher 65.7% than that of control(P〈0.05 ). Compared with control, the antibody titers of tested group at the age of 12, 19, 26, 33 and 40 days were increased by 36.6%, 40.4% (P〈0.05), 37.7%, 34.7% and 38.8%, respectively, meanwhile the SIgA in the intestinal fluid of tested group were increased by 8.59%, 10.6%, 22.8%, 68.7% and 23.2% respectively(P〈0.05). The IgG in the serum of tested group at the age of 19, 33 and 40 days were higher 17.9% (P〈0.05), 40.9% (P〈0.01) and 41.1% (P〈0.01) than that of control, respectively. Both the E-C3bR and E-ICR of tested group at the age of 19 days were higher 105.9% (P〈0.05) and 14.3% (P〉0.05)than that of control, while there was no significant difference in the above indexes between tested group and control group at the age of 26 and 33 days. In conclusion, the supplementation of compound probiotics can improve the growth performance and immune function of broilers to some degree.

    Abstract:In this test, 40 F1 black goats (Nubia goat × Longlin goat)with weight of about 16 kg were randomly divided into five groups, including group I (60% concentrate plus 40% Bachiaria hybrid powder) as a control, group Ⅱ (60% concentrate plus 10% cassava stalk powder plus 30% Bachiaria hybrid powder), group Ⅲ (60% concentrate plus 20% cassava stalk powder plus 20% Bachiaria hybrid powder) ,group IV (60% concentrate plus 30% cassava stalk powder plus I0% Bachiaria hybrid powder) and group V (60% concentrate plus 40% cassava stalk powder), and test was carried out for 60 d. Covariance analysis was used to correct the difference of initial weight of goats tested, and the effects of using cassava stalk powder to fatten the housing goats were analyzed. The results showed that the average daily gain (ADG) was no significantly different among the five groups (P 〉 0.05), while the average daily food intake (ADFI) of group I was significantly higher than that of group Ⅱ, group Ⅲ, group IV and ~ouo V (P〈0.05). In a word. it is feasible to use cassava stalk nowder to fatten the housin~ ~oat~_
    Study on Lactic Acid Fermentation and Feed Characteristics of Pomegranate Residue

    [Objective] Taking pomegranate residue as raw materials, ensiled technology, feed characteristics and dry matter digestibility were studied. [Method] Three treatments were prepared as followed: only silage (control), silage with 0.02% of lactic acid bacteria and silage with both 0.02% of lactic acid bacteria and 5% corn flour, and then packed in plastic bottles and stored a room temperature for 30 days for sampling analysis. [ Result ] The proportion of seeds and peel in residue in dry matter basis wert 18.6% and 78.6%, respectively. The crude protein and crude fat contents of whole residue and seeds were 10.84%, 10.37% and 12.70%, 13.51%, respectively. The pH values of whole treatments were less than 3.70. The organic acid was dominant by lactic acid, and butyric acid was not detected. Adding lactobacillus or adding both lactobacillus and corn flour could increase the crudt protein content and dry matter digestibility of silage significantly. [ Conclusion ] Pomegranate residue is suitable for ensiling ray materials, and no inoculation can also make quality silage. Adding lactobacillus and corn flour could increase the crude proteil content and dry matter digestibility of silage, which reached 13.03% and 71.99%, respectively.

    Chrysanthemum beer has the main ingredient of malt, and it is made through fermentation by adding hops and chrysanthemum. The chrysanthemum beer is a new product that appeared in recent years. Chrysanthemum is bitter and cold, and it can clear liver, improve eyesight, lower blood pressure, tranquilize the nerves and detoxify. Since chrysanthemum has medical function, chrysanthemum beer has more health protection effects than ordinary beer has.     In order to analyze the occurrence regularity of chronic renal failure of dog, 30 cases of dog chronic renal failure were collected from some pet hospitals in Hohhot, and the species, age, sex, occurrence time, food habits, biochemical parameters of blood and imaging examination were investigated. Results showed that hybrid dogs accounted for 37% of total cases, which was further higher than other species. The disease dogs were 2-13 years old, and the average age was 7.53; they were mainly 7-9 years old, which accounted for 53% of total disease dogs, among them 17 cases were male dogs which accounted for 57.0%, and 13 cases were female dogs which accounted for 43.0%. The occurrence time was dominant by spring and summer. The disease dogs were mainly fed with prepared-self food which was meat mostly. The disease dogs all had different degrees of anemia, and their urea nitrogen, creatinine and phosphorus increased, while Na and K decreased totally. The imaging examination result showed that the kidney of disease dog became bigger or smaller or was still normal, and the echo of kidney tissue increased.

    A clinical survey on 956 sick canines of a pat hospital in Nanning City of Guangxi was carried out in 2012, and the survey contents mainly included morbidity, onset season and age of canine parvovirus, relationship between purity of canine variety and morbidity, effects of immunity on canine parvovirus, etc. The results showed that the disease incidence of canine parvovirus was high in Nanning, reaching 14.3%; it mainly appeared in spring, late autumn and winter; canines under each age could suffer the disease, but morbidity and mortality of young canines were higher; the best way to prevent the disease is immunity.
